微波等离子体溅射诱变选育花生四烯酸高产菌及补料工艺研究

摘要
采用微波等离子(N+,15w,3min)溅射高山被孢霉T105,筛选到高产菌R254,该菌最高生物量29.3g/L,油脂11.5g/L,花生四烯酸4.20g/L,花生四烯酸产量是原出发菌株1.35倍.通过对突变株R245的代谢情况、继代稳定性以及脂肪酸组成分析,认为微波等离子溅射诱变育种是获得花生四烯酸高产菌株的有效方法.并采用补糖工艺可进一步提高其产量,花生四烯酸产量为7.43g/L,是未补糖时花生四烯酸产量的1.76倍,且为出发菌株花生四烯酸最高产量的2.40倍.
Abstract
Mutagensis R254T 105 was o btained after implanted with microwave plasma (N~+,15w,3min) sputter, in which maximum biomass, production of lipid and arachidonic acid were 29.3g/L, 11.5g/L and 4.20g/L, respectively, while the original strains T105, had 0.96 g/L of arachidonic acid production. After analysis of metabolism, subculture stabili ty and fatty acids composition in mutation R245. Implanted with ion beams is an effective method to gain Mortierella alpina containing high arachidon ic ac id concentration. After feeding batch improved further arachidonic acid producti on, maximum biomass of 40.7g/L, production of lipid of 18.5g/L and arachidonic a cid content of 7.43g/L were achieved in the mutative strain R254 in which arachidonic acid content was 1.76 times higher than that without supplementing s ugar and 2.40 times higher than that of the original strains T105.
