黄嘌呤氧化酶产生菌的离子注入诱变及其二步发酵的研究

摘要
采用剂量1.0×1012~1.0×1016?ions/cm2的N+注入产黄嘌呤氧化酶的球形节杆菌ATCC8010,研究其诱变效果.结果表明:细菌的存活曲线呈现特殊的"马鞍形",菌落形态和颜色发生了变化,且变化与产酶量有关,经过筛选获得了遗传性能稳定的高酶活突变株,使酶活较出发菌提高了43.0%.改进发酵工艺,采用二步发酵其酶得率是一步发酵的4.75倍.
Abstract
The present paper deals with biological effects of xanthine oxidase producing Arthrobacter globiformis ATCC8010 implanted by nitrogen ions with dose from 1.0×10 12 ~1.0×10 16 ions/cm 2.The results show that the survival of the strain appears "saddle shape",and the colony shapes and colors varied which related to the ability of producing xanthine oxidase. One mutant capable of producing 1.43 times as much xanthine oxidase as its original strain was obtained by selection.The optimization study on fermentation technology for the high-yield strain indicated that the production of xanthine oxidase increased by 4.75 times using two-step fermentation as compared with one-step fermentation. The yield of the xanthine oxidase is 450 units per g xanthine.
