摘要
运用均匀设计法, 对影响Rossetta(DE3)/pGEX-KG/AtGID1a表达水平的因素进行了优化, 摸索出了对可溶性蛋白进行诱导表达的最优条件, 即蛋白胨16 g/L, 酵母粉5 g/L, 葡萄糖4 g/L, IPTG终浓度0.1 mmol/L, 诱导温度23 ℃, IPTG诱导时间4 h。在此条件下, 目的蛋白的平均密度从0.042提高到0.064, 优化率为52 %。
Abstract
The factors that influenced expression level of Rossetta(DE3)/pGEX-KG/AtGID1a were optimized using uniform design. The optimization induction condition for the solubility protein expression were investigated. The results were as followed: 16 g/L peptone, 5 g/L yeast powder, 4 g/L glucose, 0.1 mmol/L IPTG, 23 ℃ induction temperature and 4 h induction time. The average density of interest protein had increased from 0.042 to 0.064. The optimization rate was 52 %.
