复合诱变被孢霉原生质体获高产γ-亚麻酸的研究
Studies on Compound Mutagenesis Method of Protoplast of Mortierella Ramanniana Obtaining High γ-Linolenic Acid Yield Strain
摘要
以拉曼被孢霉(Mortierella ramanniana)为出发菌株M5, 通过原生质体制备, 经亚硝酸和激光等进行复合诱变; 进行初筛, 经摇瓶发酵法复筛, 并测定其相关性能指标,获得一生产性能比出发菌株显著提高的突变株MF10; 其干菌体收率为46.8 g/L、油脂产率达34.6 g/L、γ-亚麻酸的产率达12.5 g/L, 分别是出发菌株的1.73倍、2.02倍和2.6倍。通过基因传代实验, 说明突变株的基因可稳定遗传。
Abstract
A mutant strain MF10 accumulating a large amount of lipid was successfully obtained when the original strain Mortierella ramanniana-M5 was treated by compound mutagenesis through the use of HNO2 and laser after its cell walls was decomposed. The content of lipid in MF10 mycelium culture in shaking flask with its all dry cell weight was up to 46.8 g/L which is 173% higher than that in the original strain,and its lipid yield 34.5 g/L and γ-Linolenic acid yield 12.5 g/L are 202% and 260% higher than its original strain.The passing generation test showed that the heredity character of mutant MF10 was stable.
