心舒宝对大鼠心肌梗死的保护作用及其药理机制

摘要
目的:探讨心舒宝对大鼠心肌梗死的保护作用及其药理机制,为临床用药提供理论依据。方法:0.25 g/mL心舒宝灌胃二周后诱导大鼠心肌梗死模型,二周后进行大鼠心肌组织的Masson’s Trichrome 染色实验;利用胰酶消化方法分离大鼠心脏原代内皮细胞,并培养于培养液。用0.005%,0.01%,0.05%的心舒宝处理细胞后,缺氧培养24 h、48 h,流式细胞仪分析其凋亡率。结果:心舒宝处理组的大鼠心肌梗死面积及其切面胶原明显小于对照组(P<0.01);0.005% 心舒宝处理的大鼠心脏原代内皮细胞在缺氧48 h后,其凋亡明显降低(P<0.01)。结论:心舒宝可能通过降低大鼠心脏原代内皮细胞的凋亡而发挥保护大鼠心肌梗死的作用。
Abstract
Objective: To investigate the the protective effect and Pharmacological mechanism of Xinshubao on myocardial infarction of rats ,and provide a theoretical basis for clinical use. Methods: The model of myocardial infarction rats were induced by 0.25 g/mL Xinshubao after two weeks,then cardiac muscle Masson’s Trichrome staining was done using trypsin digest and to isolate the rat heart primary endothelial cells,and cultured the cells in 1 640 medium. Flow cytometry analysis the apoptosis rate after Hypoxia culture 24 h,48 h of cells treated with 0.005%,0.01%,0.05% Xinshubao for 24,28 h. Results: Xinshubao treated rat myocardial infarction area and the section of collagen was significantly less than the control group (P<0.01); 0.005% of Xinshubao treated rat heart primary endothelial cells in hypoxic for 48 h,apoptosis Octreased significantly (P<0.01). Conclusion:Xinshubao may protect myocardial infarction rats by inhibiting apoptosis of rat heart primary endothelial cells.
