研究紫外线诱导的光产物FICZ(6-Formylindolo[3,2-b]carbazole)对体外培养的人皮脂腺细胞芳香烃受体(AHR)信号通路影响及相关炎症因子mRNA表达的影响, 探讨紫外线诱导痤疮发生的机制。方法:Real-time qPCR检测分析永生化的SZ95人皮脂腺细胞暴露于紫外线光产物 FICZ后, 检测AHR及其下游基因CYP1A1表达, 以及炎症基因IL-1α、IL-1β、RELA等表达。结果:FICZ组AHR表达丰度是CONTROL组(未加入FICZ组, 以下均称对照组)的1.412倍(P<0.01); FICZ组CYP1A1基因表达丰度是对照组的142.338倍(P<0.01); FICZ组IL-1α基因表达丰度是对照组的4.039倍(P<0.01); FICZ组IL-1β基因表达丰度是对照组的4.245倍(P<0.01); FICZ组RELA基因表达丰度是对照组的1.432 倍(P<0.01)。结论:紫外线光产物FICZ激活AHR信号通路, 从而影响皮脂腺细胞的炎症因子IL-1α、IL-1β、RELA等产生, 这可能是紫外线诱导痤疮发生的机制之一。
Abstract
To study the effect of FICZ (6-Formylindolo [3,2-b]carbazole) on the expression of aryl hydrocarbon receptor (AHR) signaling pathway and the expression of related inflammatory cytokines in human sebaceous gland cells in vitro, the biological effects of sebaceous gland cells. Methods: Real-time qPCR assay was used to detect the expression of AHR signaling pathway and its downstream gene CYP1A1, as well as the expression of IL-1α, IL-1β and RELA in inflammatory cells after exposure to UV light product FICZ. Results: FICZ groups of AHR expression abundance was 1.412 times to CONTROL groups and P<0.01; FICZ groups of CYP1A1 gene expression abundance was 142.338 times to CONTROL groups and P<0.01; FICZ groups of IL-1αgene expression abundance was 4.039 times to CONTROL groups and P<0.01; FICZ groups of IL-1βgene expression abundance was 4.245 times to CONTROL groups and P<0.01; FICZ groups of RELA gene expression abundance was 1.432 times to CONTROL groups and P<0.01. Conclusion: FICZ induced by UV activated the AHR signaling pathway, and then affects the production of inflammatory factors IL-1α、IL-1β and RELA in sebaceous gland cells, which may be one of the mechanisms of UV-induced acne development.
