1 引言
光谱成像技术是20世纪70年代提出的一种成像技术,通过光谱分离元件将入射的全波段/宽波段光信号分成若干波段,可同时获取探测目标的光谱特征和空间图像信息。根据光谱分辨率,可把光谱成像技术分为多光谱成像(MSI)、高光谱成像(HSI)和超广谱成像(USI),其光谱分辨率分别约为10-1λ、10-2λ、10-3λ (λ为波长),成像波段数/通道数分别约为3~10、100~200、1000~10000。本文主要介绍MSI技术,在生物医学领域,该技术最早应用于皮肤诊断[1]。目前,已有一些MSI的原理[2-3]及其在生物医学领域应用[4-8]的综述。但近年来随着材料科学及制造技术的快速发展,许多新型光学元件及光电探测器被提出,这些器件可极大地提升MSI的性能并拓展其应用范畴。本文分别从成像技术及生物医学应用等角度,综述了近年来国内外在MSI方面的研究应用现状及进展,并分析其发展趋势。
2 多光谱成像技术
MSI系统一般由照明单元、分光单元及探测单元三部分组成,其中分光单元是核心。传统的MSI系统,其分光单元有些采用分段式元件[如轮转滤光片(FW)、线性渐变滤光片(LVF)[9-10]],有些则采用可调谐元件[如液晶可调谐滤光片(LCTF)、声光可调谐滤波器(AOTF)、可调谐Fabry-Perot 滤波器[11-13]]。图1(a)和(b)分别展示了基于FW和LCTF的系统,其中FW相较于LCTF具有成本低的优点,但随着光谱频道的增加,FW的体积会急剧增大,此外,这两种方法都采用串行成像的工作模式,即每次成像只检测一个波段信息,随着检测波段数量的增加,采集时间也会相应地增多。
为突破串行成像模式的限制,研究者提出了利用复合阵列光学元件实现多通道并行成像的方法,并利用集成式元件缩小系统体积。目前实际中使用最多的复合阵列光学元件是滤光片阵列,将多波段滤光片组集成在探测器的一个像素上,以同时获取多波段信息[14]。虽然该方法能够通过并行成像方式提高成像通量,但受限于滤光片尺寸,成像波段数(n)也会相应地受限(n=4),这限制了其应用。因此,Shogenji等[15-16]提出薄平面复眼成像系统(TOMBO),如图1(c)所示,通过将微透镜阵列、滤光片阵列与光电探测器阵列一一对应,集成在同一平面基底上,能够同时获取多个波段(n=9)的光谱信息。然而,平面复眼成像系统的成像视角非常有限,当进行大范围探测时,系统体积还会相应地增大。受自然界昆虫复眼的启发,通过将微透镜阵列均匀排布在曲面基底形成曲面复眼系统,可以实现大视角探测[17-18]。先前的研究受限于加工技术,无法在曲面基底上加工精密微透镜结构,因此,Chen等提出一套真空纳米压印系统,并发展了一系列曲面制造工艺,构建了一套如图1(d)所示的多层曲面复眼MSI系统,该系统由三层结构组成:第一层是微透镜阵列,用于获取图像空间信息;第二层是填充在微透镜之间的金属薄膜,用于隔挡杂散光、防止各成像通道的信息串扰;第三层是多通道滤光片,用于分离频谱、获取图像光谱信息[17-19]。他们利用该系统分别对色盲测试卡、遥感图像、组织病理切片等进行成像,证明了该系统的分光能力及特征识别能力,并展现了其在病理研究中的应用潜力。

目前便携式是MSI系统的一个发展趋势,MSI系统多与智能手机结合使用,如图2所示,常用于移动医疗检测[20-21]。在照明单元端,Kim等[22-24]在一个白光LED前放置一套转轮滤光片,对样本依次进行单色照明成像;Kuzmina等[25]则采用多色LED阵列照明系统,每次只使用单一LED/波长进行照明。此外,为缩小系统体积,可以省略分光单元,利用频谱解调分析方法,直接从手机采集图像中分解不同的波段信息,如He等[26]通过Wiener滤波算法,将手机采集的混叠RGB图像转换成可见频谱(470~620 nm)的16个波段多光谱图像。

显微成像是MSI系统的另一个发展方向,MSI系统多与荧光显微镜相结合,用于对多色荧光标记样本进行探测[27],如图3(a)所示,针对特定标记的生物结构,选择合适的激发光源,并根据发射光谱,合理设计探测光路的分光模块,实现多光谱并行探测[28-29]。为进行体内组织检测,MSI也应用到内窥显微镜中,如图3(b)所示,在照明单元通过光纤将不同波段的激光束进行耦合,在探测单元通过分光元件如轮转滤光片进行单光谱探测[30-31]。为实现体内深层组织的实时成像,还可以把多光谱技术与光声成像融合,构建多光谱光声层析成像系统(MSOT)[32-34]。如图3(c)所示,多光谱光声层析成像的原理是使用多个波长的脉冲激光照射生物组织,同时刺激组织内多种发色团吸收电磁能以实现瞬态局部加热,触发热弹性膨胀以产生压力波,通过超声传感器探测压力波并使用断层扫描反演算法将检测信号转换为含空间信息的组织多光谱图像[35-36]。

近年来,随着材料科学和制造技术的快速发展,越来越多的新材料被用于构建用于多光谱成像的新型光学元件,以优化系统性能,如提高频谱分辨率及紧凑性等。超表面是其中一类新材料,一般是由纳米结构组成的二维阵列,凭借自身奇特的电磁特性,可以实现对不同波段光束的操控,并可用于多光谱探测。Avayu 等[37]引入了多层密集垂直结构,如图4所示,其中每一层都由不同的材料[如铜(Au)、铝(Al)、金(Ag)]制成,并且针对不同的光谱带对材料进行了优化,分别设计其直径D和间距l,基于优化结构,可以设计一款用于受激发射损耗(STED)显微成像的超薄双层透镜,该超薄双层透镜能够分别将绿光和红光聚焦,形成共焦点的完整的圆形光束和环形光束,实现单元件的光谱复用。如图4(c)所示,通过设计并制造单一光学元件,能将不同波长的光(RGB)进行聚焦,且较短波长的光聚焦在较长波长的光之前(图中z为焦距)。Khorasaninejad等[38]设计了一种具有色散效应的平面透镜。通过设计纳米结构的几何特征(长度L、宽度W、高度H)和空间分布(角度θ、横向间距Sx、纵向间距Sy)等参数,该透镜能够在同一视场内形成两张螺旋度相反的图像,实现不同手性检测。值得注意的是,传统光学元件如滤光片等一旦制造完成,则无法再动态调节其光谱透过特性,而超材料可以打破这一限制。Tang等[39]将不同大小的HgTe量子点做成分层结构,通过调节偏置电压的极性和大小,实现近红外波段与远红外波段的快速切换探测,调制频率达到100 kHz。Meng等[40]提出多层H型的石墨烯阵列,通过调节费米能量和电介质的折射率来动态地调整吸收峰的光谱位置。Duempelmann等[41]提出构建基于周期性银纳米线的可调谐等离子体滤波器,通过转动不同角度实现入射波长相移。Li等[42]在石墨烯表面构建垂直的Ge-Si纳米线,通过调节偏置电压实现响应光谱的动态调节。值得注意的是,虽然很多基于新材料或超结构的多光谱成像元件被提出,但受限于制造技术,这些元件目前仍难以大面积/规模化生产,因此目前的研究多是进行原理验证,尚未进入实际的生物医学应用。

3 生物医学应用
在生物医学的科学研究及临床应用中,人们往往希望采用对生物样本损伤少的成像方式获取足够多的目标生物信息。多光谱成像技术采用非侵入式的成像方式,而且能够从细分的光谱图像中获取有效的信息,因此在生物医学领域有广泛的应用,如病菌/活细胞识别、脑缺血检测、药物开发等[43-46]。本文从病理分析、手术引导、生物识别三个方面,介绍多光谱成像技术的最新进展。
3.1 病理分析
病理分析是现代医学的重要手段,从现代病理解剖学之父Giovanni Morgagni (1682—1771)探索疾病与器官病理变化的联系,到Rudolf Virchow(1821—1902)推广使用显微成像术以开拓微观病理学,人们一直致力于将病理解剖发展成一门精确的科学[47-48]。然而,传统病理研究需要先把组织制成薄片,通过外部介入方式(如HE染色)来增强对比度,该方法并不适用于体内检测,而且只提供少量化学信息。此外,显微成像的探测范围一般为可见光波段(300~780 nm),这限制了某些微观结构的观测。而MSI技术不仅可以获取传统RGB三通道数据,还能进一步细分,获取更多窄带波谱信息,甚至可用于获取超越可见光波段的信息(如紫外或红外波段),捕获肉眼无法分辨的隐藏信息。此外,MSI技术不仅可用于染色组织分析,也可应用于未染色组织的分析。这些优点使MSI技术在病理分析和研究中有着广泛的应用[49-50]。
以皮肤组织为例,基于生物分子对近红外光谱的吸收差异,Spreinat等[51]构建一套近红外成像仪器(900~1500 nm),如图5(a)所示,采用宽场透射式照明分析固定的皮肤组织切片,并通过InGaAs探测器采集图像信息,对多光谱图像进行分析以区分健康与病变组织(如黑色素瘤、结节性基底细胞癌、鳞状细胞癌等)。此外,利用MSOT成像深度大的特点,还可以在体探测皮肤组织的动态过程。如图5(b)所示,Schwarz等[52]将多光谱纳秒激光脉冲耦合到光纤束中并激发皮肤组织,通过超声换能器采集空间深度信息,通过光电二极管收集每个激光脉冲的反射光,解调多光谱数据集,获取表皮中黑色素含量和血液氧合值,还可以通过定量分析黑色素的直径、颜色和分布边界的非对称性与不规则性,对皮肤病变进行预判[53]。He等[54]将智能手机与手电筒相结合,构建一套可用于实时监测的便携式光谱成像系统,通过Wiener算法将手机内置RGB相机的采集图像转化为16波段的多光谱图像(470~620 nm),提取包含血红蛋白与黑色素等特定发色团的分布特征,监测心动周期和血管阻塞下的皮肤血液动力学活动。

目前在病理研究中,多光谱技术最常应用于肿瘤检测。Wu等[55]发展了两种近红外激活的光声探针(C1X-OR1 和C2X-OR2),并采用MSOT进行疾病诊断和定位病灶,其中C1X-OR1用于定位小鼠肝损伤位点,C2X-OR2用于定位转移性肿瘤。Jo等[56]发展了一种肿瘤靶向、pH传感的纳米探针,结合MSOT成像技术,能够同时测量肿瘤微环境pH值以及血液动力学特性。而Habibalahi等[57]利用多光谱自发荧光成像并结合机器学习方法,能够在无标记的条件下识别良性组织与肿瘤组织并进行边界检测。
3.2 手术引导
由于MSI技术能够识别不同的生物组织信息,因此在手术过程中利用多光谱图像引导、协助医生定位病灶具有较大的应用价值。借助特异性荧光示踪剂,多光谱成像有可能解决病变组织边界模糊导致的难以识别、切除不彻底等问题,以提高手术精度[58]。如图6所示,Goh等[59]利用多光谱光声层析成像,提高乳腺癌肿瘤边缘识别的准确率。Hu等[60-61]发展了一套融合可见光波段及近红外I、II区(400~650、700~900、1000~1700 nm)的多光谱探测成像仪器,用于引导、协助进行原发性和转移性肝肿瘤的手术切除,其中可见光波谱用于采集组织解剖信息,近红外II区用于识别、标记肿瘤荧光信息。与传统单一的近红外I区检测相比,该仪器可实现更高的肿瘤检测灵敏度(100% vs. 90.6%)、更高的肿瘤与正常组织信号比(5.33 vs. 1.45)以及更高的肿瘤检出率(56.41% vs. 46.15%)。Qiu等[62]研制一套内窥MSI系统,能快速扫描整个食道内膜,通过分析上皮细胞的散射光来检测和识别肉眼无法判断的增生部位,检测准确率能达到90%。Schols等[63]利用MSI技术(440~1830 nm),能够在结肠样本中区分出5种不同类型的组织(即肠系膜脂肪、血管、结肠、输尿管和结肠肿瘤),在腹腔镜结直肠手术中进行图像引导。此外,MSI技术还可用于识别荧光标记的不同类型的结构,用于临床图像引导。van Willigen等[64]利用多光谱腹腔荧光显微成像系统,依次采集三种发射光谱互补的荧光示踪剂(荧光素、Cy5和ICG)数据,以在临床手术中区分正常与病变组织。Behrooz等[65]利用Solaris多光谱成像系统,通过同步实时背景去除方法,在手术照明的环境下以nmol量级的灵敏度对弱荧光信号进行采集,同时通过多光谱图像解调,区分目标荧光信号与自发荧光信号,实现开放环境下的术中图像引导。

3.3 生物识别
生物识别技术是通过获取人们的生物特征,包括生理特征(脸像、指纹、掌纹、虹膜等)和行为特征(笔迹、声音、步态等)来区分不同个体的技术。生物识别技术已广泛应用于电子支付、门禁检入、社会治安等领域,成为日常生活的重要组成。MSI技术能够采用非接触的方式快速、准确地获取并识别不同生理特征,是生物识别技术的一种有效方法。对于用于生物识别的MSI系统,如何针对不同的生物特征选择合适的成像波谱和波段数,是提高识别准确性及效率的关键。下文将针对不同的生理特征,逐一介绍成像参数设计。
虹膜是围绕瞳孔的彩色环,具有很高的排他性和防伪性,因此被广泛用于安全领域。传统成像使用近红外波段(700~850 nm)进行虹膜识别。如图7(a)所示,Chen等[66]使用可见光(480 nm)及近红外(860 nm)波段,识别结膜血管和虹膜纹理,识别准确率约为88%。而Vilaseca等[67]则利用可见光波段(400~700 nm)多光谱成像系统识别虹膜的光谱反射率及颜色,通过实验研究,使用三波段(RGB)的识别准确率可达到99%。Gong等[68-70]研制了一套三波段(700,780,850 nm)的便携式双目MSI系统,基于该系统采集一系列多光谱虹膜数据集,并利用一维log-Gabor小波滤波器进行特征提取,以验证该系统的有效性。

指纹和掌纹也是两种独特并且可靠的生理特征,在识别过程中,人们关注的是内皮纹理信息,利用皮肤对不同波谱的吸收和反射差异,结合多光谱成像可获取不同皮肤组织深度的特征信息,提高识别的准确性。目前研究的重点在于如何发展多光谱指纹/掌纹数据的分析方法并提高识别的准确率[71-76]。传统识别系统多采用白光作为照明光源,识别精度和防欺骗能力受限,因此,Zhang等[71-72]提出一种k-均值聚类算法对520~1050 nm的掌纹光谱数据进行分析,以确定特征波段的最佳数量。实验结果表明三个特征带足以表征掌纹特征,基于三个特征带的识别准确度可比拟基于四个特征带的识别准确度且优于基于两个特征带的识别准确度;利用该研究结果指导构建一套在线掌纹识别系统,并提出一种基于集合理论的分数级融合方法,以减小不同频谱之间的重叠效应。在Zhang等的研究基础上,Gumaei等[73]提出一种基于自动编码(AE)和正则化极限学习器(RELM)的分析方法,通过AE降低提取特征维数,并将简化后的特征导入RELM进行分类,这样能够有效减少掌纹特征数量并加快识别,并且不会降低分类的准确性。Xu等[74]提出一种四元数模型的多光谱图像分析方法,如图7(b)所示,该方法能有效避免某些光谱信息在分级匹配融合过程中丢失,识别准确率达到98.83%。
除了掌纹,手掌血管脉络也是一种独有的生理特征,因为其难以被伪造,近年来也逐渐被用于身份识别领域[77]。近红外是静脉成像的常用波段,为确定最佳窗口,Paquit等[77]提出一种优化方法,在495~945 nm的光谱范围以10 nm为步进,均匀照明皮肤并采集了手上不同部位的图像(获取46张图像),采用主成分分析和线性判别分析方法对多光谱图像数据进行降维处理,再使用Stegers算法检测静脉及其宽度,从中筛选出最真实的静脉,对应的照明光谱则为优化窗口。为提高静脉识别准确率,Stanuch等[78]构建了一套融合近红外和紫外的MSI系统,其中近红外窗口用于手掌静脉成像,紫外窗口用于获取掌纹信息,并使用深度卷积神经网络进行图像处理,系统识别静脉的准确率达到99.5%。此外,有效的静脉可视化对于各种临床护理有重要意义,如进行皮下注射时能方便针头插入[79-80]。Song等[79]构建一套基于智能手机的反射式成像系统,采用多光谱Wiener估算方法获取反射光谱,并将其与光谱仪的采集数据进行比对,最终实现手掌血管的可视化。
面部识别主要捕获一些面部物理特征,如眼窝、颧骨区周围以及嘴部外侧轮廓等,与其他生理特征的识别相比,面部识别具有成本低的优点[81-82]。Singh等[83]采用可见光与红外光融合的多光谱成像方法提高脸部识别的性能,如图7(c)所示,通过使用二维log-Gabor小波滤波器,从离散小波变换(DWT)融合的图像中提取幅值和相位信息,并通过自适应支持向量机(SVM)学习算法智能地选择幅值和相位特征,识别的准确率为97.14%。Zheng[84]针对包含可见光波段及远红外波段的多光谱人脸数据集,在Gabor小波变换和汉明距离的框架下,提出一种基于方向的人脸识别算法,与三种经典的算法(主成分分析、线性判别分析、弹性束图匹配)相比,所提算法具有更高的识别准确率(97.92%)。
4 结束语
多光谱成像技术的发展趋势主要有两个方面:一方面,通过将传统多光谱成像技术与新型光电材料相结合,优化系统的光谱探测范围、光谱分辨率、系统体积等性能指标,提高系统的普适性;另一方面,结合应用场景,多光谱成像系统分别往便携式系统和显微成像系统发展。
由于采用非侵入式的成像方式并能获取多维生物信息,多光谱成像在生物医学领域有着广泛的应用。除本文综述的病理分析、手术引导和生物识别等三个方向,多光谱成像还在脑科学研究、活体成像等不同领域发挥重要作用。作者认为基础研究和临床应用是多光谱成像应用的两个重要发展方向。
