1 引言
光学方法是继传统超声成像、核磁共振成像和X射线断层成像等技术之后最具潜力的生物医学成像手段。随着生物医学成像研究的深入及临床应用的不断发展,针对组织细胞的成像需要满足微米量级的分辨率、足够的成像深度和合适的信噪比(SNR)及对比度,同时还需要考虑成像的安全性和实时性。
光学相干层析成像技术(OCT)作为一种高分辨、非侵入性、高信噪比的断层成像技术,在生物医学和材料检测等领域被广泛地研究和应用[1-7]。OCT基于外差探测原理,使用低相干宽带光源,微米量级的相干门可以有效抑制杂散光,适用于高散射样品的成像。光学相干层析显微成像(OCM)技术作为OCT的改进技术,继承OCT高轴向分辨、高信噪比、高速成像等优势[8-12],使用高数值孔径的物镜,显著提高成像的横向分辨率,目前最高横向分辨率可达亚微米量级[13]。
OCM技术作为一种光学成像技术,无需标记的非侵入特性保证了无损害成像。相干门和共聚焦配置的结合可以显著抑制杂散光,保证深层组织成像下的足够信噪比。OCM并行探测结合优化的系统和数据处理算法,能够实现合适深度范围内样品的实时成像,因此,OCM技术在生物医学、材料检测等领域的应用受到国内外研究者们的广泛关注[14-19]。虽然OCM技术作为光学显微术的一种,除了具有横向分辨率的优势外,人们早就意识到其在深层组织成像上的优势[14],但其在应用上的推广却远没有达到OCT的水平,亟待引起各界的高度重视。
本文将首先介绍OCM技术的基本原理和实现方案,然后详细阐述OCM技术的研究进展和应用。针对OCM技术中如何实现超高分辨成像、焦深限制成像深度等问题,总结了目前该研究领域一些先进的OCM技术。OCM在实际应用中具有广泛性、可靠性和灵活性。
2 OCM基本原理
OCM技术作为高数值孔径物镜条件下的OCT,最大的优势在于能克服OCT横向分辨率低的缺点。不同于传统光学显微术中的强度探测,OCM采用基于相干门的干涉探测方法,兼具多次散射光的抑制及单次散射光的增强功能,可显著提高组织成像的对比度和信噪比,能够对更深层的组织样品进行光学成像。
依照扫描方式区分,OCM主要有点扫描OCM(PS-OCM)和全场OCM(FF-OCM)两种实现方式。本节将分别介绍这两种OCM的原理和OCM系统的主要性能参数。
2.1 点扫描OCM
共聚焦光学显微术是目前广泛应用的可以实现轴向层析成像的技术,它由Marvin[20]于19世纪50年代后期首次提出。该技术中,点光源发散光由物镜聚焦到要成像的样品表面或内部,聚焦到焦点位置的光经反射后可以通过探测器前设置的小孔而被探测到,离焦位置处反射的光则被小孔显著抑制。共聚焦的设计能够减少离焦位置散射光引起的成像模糊,进而提高成像分辨率和信噪比[21]。
1991年,Huang等[22]首次提出OCT,OCT基于低相干干涉(LCI)技术,在单点深度信息探测的基础上加入X-Y二维扫描,实现对样品的三维成像。Izatt等[23]首次提出时域OCM(TD-OCM),将共聚焦模块加入OCT系统的样品臂中,如

在TD-OCM中,轴向移动参考臂反射镜用于单点深度信息的探测。如

在TD-OCM中移动参考镜的过程中,由于存在相干长度外γ(z)快速衰减为约等于0的限制,只有两臂近似相等时,才有(4)式所示的干涉信号产生。这样通过参考镜的一次快速轴向扫描,根据探测干涉信号的强度和位置,就可以得到样品内部各散射界面的位置和散射率的大小。通过轴向探测加上光点在样品表面的横向二维扫描,系统就能实现三维成像。
1995年,Fercher等[24]首次提出频域OCT(FD-OCT)的概念,频域OCM(FD-OCM)技术也得以发展。和TD-OCM相比,FD-OCM不仅在成像速度上有极大的提升,成像灵敏度也成倍增大[25-27]。

FD-OCM的实现方式有谱域OCM(SD-OCM)和扫频OCM(SS-OCM)两种。保持FD-OCM中参考平面镜固定不动,SD-OCM利用宽谱光源和光谱仪的组合,SS-OCM利用扫频光源和点探测器的组合,采集到的光谱域中的干涉信号为

从(6)式可以看到,FD-OCM可以并行获得样品单点整个探测深度的信息,轴向并行探测结合CCD(CMOS)技术和扫频光源,使得FD-OCM成像速度得以大大提升。同时,参考镜的固定也减少了机械移动带来的相位噪声,提高了成像的信噪比和对比度。目前为止,FD-OCM逐渐替代了TD-OCM,成为PS-OCM主要的实现方式。
2.2 全场OCM
1998年,Beaurepaire等[28]首次提出FF-OCM,系统示意图如

可以看到,FF-OCM系统干涉信号的产生和接收的核心在于低空间相干光全场照明以及干涉信号相位的调制和解调。另一种常见的FF-OCM系统使用压电陶瓷晶体(PZT)实现相位简谐调制[35]。

2.3 OCM系统参数
结合上面的原理讨论可知,相干门大小决定OCM系统的轴向分辨能力,轴向分辨率为

1998年,在Beaurepaire等[28]首次报道FF-OCM后,多个课题组在FF-OCM技术上进行了更加广泛深入的研究。2002年,Beaurepaire课题组[29]证明在FF-OCM中,当显微物镜数值孔径较大时,很窄的聚焦光点轴向分布也会决定系统的轴向分辨率,即δzobj=
上文已经对成像分辨率进行了分析,OCM系统的焦深限制也需要被重点分析,其他参数详见参考文献[ 23]。由于使用高数值孔径的物镜,光束被高度聚焦,因此焦深限制为
光点在焦点以外会快速扩散,导致散射信号无法或很少返回,严重限制成像深度。在OCT中,使用小数值孔径(<0.1)的物镜可以保证1~2 mm的探测深度,而OCM由于使用高数值孔径(0.7~1.2)的物镜,探测深度被限制到数百微米,严重限制OCM系统在实际中的应用。
3 超高分辨OCM研究进展
OCM作为一种光学显微技术,实现高分辨、高信噪比成像一直是贯穿OCM技术发展历程的研究方向,相关的研究进展也表明OCM能够应用于生物、医学、材料检测等诸多领域。
3.1 超高分辨OCM技术的发展
初期的OCM系统大多使用LED、SLD等作为光源,带宽为几十纳米,因此轴向分辨率只能达到10~20 μm。1997年,Schmitt等[36]将1240 nm和1300 nm中心波长、40 nm带宽的两个LED耦合到一起,将单个LED下系统的10.8 μm轴向分辨率提高到7.2 μm。但是多个LED光源耦合要求的连续多个不同波段LED光源难以获得,更大的带宽实际上不易实现。超连续谱激光器技术的发展和成熟,可以提供200 nm带宽的低相干光源,使得OCM可以实现3~5 μm的轴向分辨率。2003年,Aguirre等[37]报道了使用飞秒钛宝石激光器(λ0≈800 nm,FWHM约为200 nm)可实现3 μm的轴向分辨率。2015年,本课题组[38]报道的谱域系统使用超连续谱激光光源(λ0≈665 nm,FWHM约为230 nm)可实现0.9 μm的超高轴向分辨率。

亚微米量级的成像分辨率可以实现对生物组织细胞甚至亚细胞结构的成像。如

通过浸水或浸油可以将物镜的数值孔径提高到1以上,从而进一步提高FF-OCM系统的分辨率。2015年, Dubois课题组[13]报道了使用数值孔径为1.2的水浸物镜,FF-OCM系统实现了0.5 μm(水中)的横、轴向分辨率,成像信噪比为71 dB,成像深度约为100 μm。
使用短波段、宽带光源和高倍物镜的FD-OCM系统也能获得亚微米量级的分辨能力,实现生物组织亚细胞量级成像。2017年,Marchand课题组[39]报道的FD-OCM系统实现了0.7 μm和0.4 μm的轴向和横向分辨率,系统所用光源中心波长为647 nm、带宽为246 nm,物镜放大倍数为40×(有效数值孔径大小为0.76),成像深度约为40 μm。
3.2 OCM系统的多次散射抑制和色散补偿
OCM系统在实际应用中要获得高分辨、高信噪比的成像,除了要考虑光源、物镜、探测器等器件的性能,对多次散射抑制和色散补偿也要引起重视。
在高散射样品中,OCM探测样品的背向散射光可能会经过多次散射返回并被探测,多次散射的过程中会引入随机的相位。多次散射引起的时延会展宽探测信号的PSF,引入的背景噪声会减小成像的对比度[40-41]。这种散斑噪声在OCT中普遍存在,如何抑制散斑噪声也一直被多个课题组深入研究[42-45]。在PS-OCM中,使用相干门和高倍物镜下的共聚焦配置,可以有效限制多次散射,避免散斑噪声对成像的影响。而在FF-OCM中使用面阵探测器实现宽场探测,缺少共聚焦配置使得多次散射后背向散射光偏离入射光束,引起探测器相邻像素之间的光学串扰[46]。
对于FF-OCM中多次散射引起的光学串扰问题,多个课题组使用低空间相干光源来抑制串扰[47-48]。2019年,Wojtkowski课题组[49]提出用可变形镜对低空间相干光附加随机的相位,使得照明样品的光空间不相干,实现了无光学串扰的散射样品成像。

但是,尽管光源的空间非相干能够减小光学串扰造成的分辨率损失,但是本质上并没有消除光学串扰引起的噪声,因此不能有效提高信噪比。到目前为止,FF-OCM依然只适合浅层低散射样品的成像,深层生物组织成像的OCM技术将在第4节讨论。
另外一个显著影响OCM系统分辨率的因素来自参考臂和样品臂两臂间的色散失配。超高分辨OCM系统为了实现高轴向分辨率,使用超宽谱宽(FWHM大于200 nm)的光源,非共路设计带来的样品臂和参考臂色散失配会严重降低分辨率[50],同时,对一定深度的样品进行成像时,色散失配会随深度的增大而增大。2015年,Federici等[13]研究了超高分辨OCM系统的轴向分辨率和探测深度、光源带宽的关系,以人表皮组织为样品的实验结果如

因此,合适的色散补偿方法是必须要考虑的,在参考臂中加一块玻璃板,通过调节玻璃板的厚度或角度可实现近似的色散补偿[50]。2001年,Fercher等[51]提出数值色散补偿方法,在后期数据处理过程中,将干涉信号乘上一个只和波长有关的补偿相位,通过迭代调整补偿相位使得PSF最窄。2017年,Lee等[52]报道了在进行数值色散补偿前对干涉光谱进行加窗Wiener滤波可以进一步锐化PSF函数。2017年,Pan等[53]报道了一种依赖深度的色散补偿算法,对于不同的深度迭代计算色散补偿量,可实现全深度的色散补偿。
4 突破焦深限制的OCM技术研究
超高分辨OCM技术应用于生物医学领域,能够对一定深度的生物组织实现探测成像。从第2节原理讨论可知,OCM使用高数值孔径的显微物镜实现了高横向分辨率,但随之带来的焦深减小会严重限制成像深度。FF-OCM使用宽场照明和面阵成像的方式可以避免焦深限制,但是缺少共聚焦配置,不能有效抑制杂散光,因此PS-OCM更适合深度成像。
在PS-OCM中要想获得一定深度区域的样品显微图像,最常用的方法是沿轴向移动样品,重复获得不同深度区域的图像,再通过合适的算法进行拼接[54-56],然而这种动态聚焦成像方式不仅会严重降低成像的速度,同时多次移动物品和图像拼接也会带来额外的误差。相关课题组通过分析光场分布去卷积的算法实现数值变焦[57-59],经过数据后处理,得到样品成像在较深区域具有较高的信噪比,但这在实际高散射生物组织成像中是不易保证的。Yamanaka等[60-61]使用1700 nm波段的光源,在TD-OCM系统和FD-OCM系统中实现了对猪甲状腺1.2 mm和1.8 mm的探测深度,但是和常用的800 nm探测波段相比,使用1700 nm探测波段的系统难以获得亚微米量级的超高分辨率。而使用类贝塞尔函数分布光束来实现焦深扩展和基于液体物镜自动变焦的Gabor域OCM(GD-OCM)是被广泛研究的2种技术。
4.1 焦深扩展OCM
2002年,Ding等[62]提出用锥镜产生贝塞尔光束照明样品实现OCT系统的焦深扩展。高斯光束经过锥镜后在焦平面会形成贝塞尔函数分布的光场模式,在远场会形成圆环形分布的光束。焦平面贝塞尔光场分布的主瓣在轴向会保持很长一段距离而不横向扩散,这种特性使得此光束用于PS-OCM探测时可以在保证焦深扩展的同时不损失高分辨能力。但是,将锥镜聚焦的光点直接用作探测光的问题在于光束主瓣能量占比很低(1/e2宽度内约为5%),而理想高斯光束主瓣1/e2宽度内能量占比为86%,这种情况下要取得较高的信噪比,需要较强的入射光强,这在生物组织成像尤其是临床应用上有很大的限制。2007年,Liu等[63]利用二相空间滤波器代替锥镜产生类似贝塞尔分布模式的光束,其主瓣1/e2宽度内能量占比提高到了28%。2006年, Leitgeb等[64]使用双透镜将锥镜后远场环状光束进行了准直再聚焦,散射光为类高斯分布,主瓣能量占比得到了明显提升。后续研究表明这种照明方式可以实现照明光和探测光分离的暗场照明[65-67]。

4.2 基于物镜变焦的OCM技术
2010年,Rolland课题组[69]提出一种基于变焦物镜的GD-OCM技术。GD-OCM在硬件方面使用液体变焦物镜,通过改变施加电压改变焦距实现2 mm深度的成像,在算法方面利用Gabor变换实现图像在轴向的拼接,保证在整个成像范围内保持高分辨率。图像拼接的核心思想是对不同深度处获得的图像沿轴向进行加窗处理并进行叠加,具体数据处理原理可参考文献[
70]。
GD-OCM对非洲蛙蝌蚪成像结果如
焦深扩展、GD-OCM等技术突破了PS-OCM中焦深对成像深度的限制,进而可以充分发挥PS-OCM技术深度域并行采集的优势,性能得到提升的OCM系统对深层样品的实时三维成像将成为可能。硬件上,CCD(CMOS)技术和改进的扫频光源能直接提高数据采集的速度。2011年,An等[73]利用双相机实现0.5 MHz的线扫描频率。垂直腔面发射激光器(VCSEL)作为扫频光源的SS-OCM系统,线扫描频率可达到1.2 MHz [74-76],FF-OCM等效线扫描频率达到1.5 MHz [77]。同时,传统的插值、快速傅里叶变换(FFT)算法需要耗费大量的资源和时间,通过优化信号处理算法来提高数据处理速度也一直被广泛研究[77-81]。2013年,Podoleanu等[82-83]报道了一种主从干涉术(MSI),先用平面镜作为样品产生模板矩阵,再将实际样品信号和模板进行卷积得到同FFT类似的结果,但该方法能大大减轻计算负担。后续的研究引入复数主从干涉术(CMSI)的算法,改善了MSI计算引起的分辨率下降的问题[84-85]。2016年,Xue课题组[86]将光计算引入OCT中,实现了109 pixel/s的数据处理速度。2019年,采用优化的光计算OCT系统实现了5×109 pixel/s的数据处理速度,分辨率得到明显的提升[87]。可以预见,实时三维成像OCT将趋于成熟,因此将相关技术应用于OCM技术中亟需得到深入的研究和跟进。

5 OCM应用研究
OCM作为一种光学显微技术,实现高分辨、高信噪比、实时成像一直是贯穿OCM技术发展历程的研究方向。
5.1 结构成像在疾病诊断中的应用
OCM具有微米量级的分辨率,可以对生物组织细胞的结构进行超高分辨成像,无标记、高信噪比的结构成像可以直接反映一些疾病的病理性特征,因此,OCM技术在眼科、皮肤科、肿瘤学等领域得到了广泛的研究[13,91-96]。
眼科是OCT应用最为广泛的领域,经过20多年相关技术的发展和完善,OCT已经成为眼科诸如青光眼、黄斑变性、糖尿病黄斑水肿、多发性硬化等疾病诊断的“金标准”[97-100]。OCM在牺牲一定成像深度下拥有比OCT更高的横向分辨能力,能够对眼局部结构进行成像,高分辨成像可以对病变组织特征进行更精准的诊断。
除了在生物医学成像上的广泛应用,OCM还可以和多种其他成像技术结合。利用荧光成像、光声成像等可以揭示和补充OCM,进而得到组织细胞的空间结构信息,为相关疾病提供了更精准的诊断[39, 101-103]。

5.2 基于动态测量的功能成像
运动是活体生物组织细胞始终存在的特征,高速实时成像的OCM技术利用对组织细胞的长时间探测,获取其在不同时间下形态和位置的变化,以反映其新陈代谢的过程。因此,除了能对生物组织样品的空间信息结构进行成像,基于动态测量的OCM技术还可以获取血流、组织细胞活动等功能信息[108-110]。

6 总结和展望
详细介绍了OCM技术的原理和实现方式,重点总结了超高分辨OCM技术的研究进展和应用,阐述了突破探测深度限制的相关技术。在OCM技术两大实现方式中,FF-OCM技术用普通白光光源进行全场照明和探测,高倍物镜下能实现亚微米量级的超高分辨显微成像,尤其适合浅层样品成像;PS-OCM技术使用相干门和共聚焦配置,能显著抑制杂散光,在深层组织成像上有独特的优势。而焦深扩展、GD-OCM等技术能克服OCM中焦深限制探测深度的瓶颈。
OCM技术作为一种新兴的光学成像方法,依然存在很大的提升空间。随着CCD(CMOS)、扫频光源等核心器件扫描速度的提升以及数据处理算法的优化,实现大视场实时成像的OCM技术还有待进一步完善。除此之外,基于动态探测的OCM技术可以获得血流、组织活动等功能信息,其与空间显微结构成像的结合能为一些疾病的诊断标准和治疗提供有力的辅助手段。
