可见光响应的CdTe/TiO2复合纳米颗粒体外PDT灭活HL60细胞的研究

摘要
文章采用溶胶凝胶法制备核壳CdTe/TiO2复合纳米颗粒, 探讨了该复合纳米颗粒体外PDT对HL60细胞的灭活作用。通过扫描电镜(TEM)、X射线光电子衍射仪(XPS)对CdTe/TiO2进行表征。文中, 用紫外可见光吸收光谱(UV-vis)测得尺寸为2-5 nm的CdTe QDs吸收峰为460 nm。研究表明, CdTe/TiO2复合纳米颗粒尺寸在80 nm左右, 其吸收光谱相较于TiO2的光响应区拓展至可见光区。将CdTe/TiO2与HL60细胞进行共同孵育, 采用CCK-8法研究了其在暗室条件下细胞的生长情况和浓度对细胞相对存活率的影响以及在不同浓度的CdTe/TiO2复合纳米颗粒PDT后的细胞活性。实验结果表明: 在共同孵育16 h后CdTe/TiO2对HL60细胞的毒性最强, 10~320 μg/mL浓度的CdTe/TiO2样品对HL60细胞均具有较强的灭活作用。当添加CdTe/TiO2样品浓度为320 μg/mL时, 光照1 h后PDT灭活效率达到87.7%。
Abstract
CdTe/TiO2 composites nanoparticles were prepared by hydrothermal assisted sol-gel technique, which were used to do a study on PDT inactivation of HL60 cells in vitro in this paper. CdTe/TiO2 was characterized using transmission electron microscope (TEM), X-ray photoelectron spectroscopy (XPS) and FS technique. CdTe QDs with a 460 nm absorption peak were obtained by use of UV-vis adsorption spectra with an estimated size of 2-5 nm. The results indicated that the particle size of CdTe/TiO2 composite nanoparticles is about 80 nm, and its absorption spectrum showed that light response region was expanded into the visible region compared with TiO2. When HL60 cells were cultured with CdTe/TiO2, we studied the proliferation effect on HL60 cells and the effects of final CdTe/TiO2 on the relative survival of HL60 cells in the darkroom conditions, as well as cell activity after PDT with CdTe/TiO2 in different concentrations. The experimental results showed that highest cytotoxicity of CdTe/TiO2 by co-culture with HL60 cells after 16 hours, and the CdTe/TiO2 sample had an obvious inactivation effect on HL60 cells when its final concentration was between 10 μg/mL and 320 μg/mL. With one hour light irradiation, the efficiency of PDT inactivation reached 87.7%, when the concentration of CdTe/TiO2 sample is 320 μg/mL.
