BioTechniques人物专访:谢晓亮
谢晓亮(Sunney Xie)这个名字大家一定都不陌生。作为单分子研究领域的前沿科学家,谢晓亮教授大幅改良了单分子荧光显微镜技术,在相干拉曼显微成像技术(CARS、SRS)等方面成果斐然。近年来,他又在单细胞测序技术上取得突破,发表了不少成果。鉴于此,《BioTechniques》杂志2014年4月对他进行了专访,了解促使他成功的动力。在被问及当年如何确定研究方向时,谢晓亮提到了他在哈佛大学所上的一门课,Richard Losick的分子生物学课程。上课时,Losick以动画演示了聚合酶和核糖体如何工作。看到这些,谢晓亮想,总有一天他们能将活细胞中的那些过程制作成实时的电影。这个想法促使他们开发出观察和研究单细胞基因表达所需的技术。对于他的科学贡献,谢晓亮谈到了他早期的工作。他们最早开展室温下的单分子荧光成像和光谱分析。在首个实验中,他们应用这项技术来研究胆固醇氧化酶,这种酶在氧化时发荧光,但在还原时不发。在酶促循环中,荧光开启和关闭,他们追踪了这个循环。他们发现了一个现象,酶努力工作一段时间,然后放慢,但米氏方程即使在单分子水平也成立。在新技术方面,他们开发了CARS和SRS显微镜。这些作为无标记的方法,让研究人员能够对活体组织中的DNA、蛋白质、小分子代谢物或药物分子进行成像,且灵敏度比之前的方法高很多。此后,他们开始研究单细胞中的基因表达,对蛋白质和mRNA产生进行成像,每次一个分子。通过这种方式,他们能够定量地描述活细胞中的这些过程。谢晓亮谈到,他最新的研究方向是单细胞基因组学。活细胞中的单个DNA分子通过获得拷贝数变异和SNP而不断变化。不同细胞中的活性无法同步,因此必须在单细胞的基础上研究异质性。为了实现这一点,他们开发出了无偏倚的全基因组扩增方法,称为MALBAC。之后,他们利用MALBAC创建出一种新的体外受精(IVF)过程,目前正在中国进行临床试验。他们对两个极体细胞进行高通量测序。通过这些序列,他们可推断出受精卵是否可用。这项试验正在进行当中,但到目前为止,这种方法明显提高了试管婴儿的成功率。它还能够避免父母任一方与孟德尔疾病相关的点突变。同时,他们也在研究de novo突变,特别是那些导致癌症的突变。谢晓亮表示,MALBAC擅于检测这些突变,但并非收官之作。他们正在开发更好的方法来发现新突变。对于职业生涯中的亮点,谢教授认为是在临床试验中帮助患者。他回忆道,一位患者来找他。那位患者从父亲身上遗传了一个点突变,在10岁那年患上骨癌。他不希望他的孩子也像他一样。当确定怀孕的那一刻,大家都很兴奋,相信宝宝会是健康的。谢晓亮目前任哈佛大学化学与生物系终身教授、北京大学生物动态光学成像中心(BIOPIC)主任。来源:ebiotrade

Frontiers of OptoelectronicsOctober 2013Preparation, structure and properties of fluorescent nano-CdTe/poly (1, 4-butanediol-citrate) bioelastomer nanocomposite in-situ dispersion techniqueLi Jiang, Aimiao Qin, Kunpeng Jiang, Lei Liao, Xiulan Wu, Chaojian WuAbstractHydrophilic photoluminescent CdTe/poly (1, 4-butanediol-citrate) (PBC) bioelastomer nanocomposite was successfully synthesized by a two-step method and characterized by X-ray diffraction (XRD), Fourier transform infrared (FT-IR) spectroscopy, Uv-vis spectroscopy, photoluminescence (PL) spectroscopy and scanning electron microscope (SEM). The differential scanning calorimetry analysis shows that the bioelastomer nanocomposites with different mass fractions of CdTe have low glass-transition temperature, which indicates that they possess elastic property in the range from room temperature to the expected applied temperature (37°C). The measurements of the hydrophilicity, in vitro degradation and PL show that the nanocomposite has good hydrophilicity, degradation and high fluorescence properties.

The applications of independent component analysis (ICA), an efficient higher-order statistics (HOS) based blind source separation (BSS) technique, are introduced for wireless MIMO systems, along with some of the important issues surrounding them. The ICA based blind/semi-blind frequency-domain equalization schemes are proposed for wireless MIMO channels, and ICA is also used for blind/semi-blind compensation for in-phase/quadrature (I/Q) imbalance and carrier frequency offset (CFO). Simulation and measurement results show that the ICA based equalization methods can achieve a performance that is much better than those of the previous methods, and close to the case with perfect channel state information (CSI), no I/Q imbalance and no CFO. BiographyZHU Xu received the BEng degree (with first class honours) in 1999 from the Department of Electronics and Information Engineering, Huazhong University of Science and Technology, Wuhan, China, and the PhD degree in 2003 from the Department of Electrical and Electronic Engineering, the Hong Kong University of Science and Technology, Hong Kong. Since May 2003, she has been with the Department of Electrical Engineering and Electronics, the University of Liverpool, Liverpool, UK, where she is currently a Senior Lecturer. Dr. Zhu has over 100 refereed publications in highly ranked international journals and conference proceedings, in the broad area of communications and signal processing. Her research interests include wireless MIMO systems, equalization, OFDM techniques, resource allocation, cooperative communications, cross-layer design, cognitive radio, ultra-wideband radio and smart grid communications. Dr. Zhu is a senior member of IEEE and a member of IET. She is an Associate Editor for the IEEE Transactions on Wireless Communications, and has served as a Guest Editor for the ICSAI 2012 Special Issue in Computer Science and Information Systems. She was Vice Chair of the 2006 and 2008 ICARN International Workshops, Program Chair of the ICSAI 2012, and Publication Chair of the IEEE IUCC-2012. Dr. Zhu has received research grants of over £850k since 2006, including £600k as the Principle Investigator, from various funding bodies such as EPSRC, KTP and EU. Since 2006, she has graduated 7 PhD students and over 30 MSc students as the first supervisor, and has supervised 6 post-doc researchers successfully.
