Femtosecond laser direct writing processing of SERS substrates and applications

Overview: Surface-Enhanced Raman spectroscopy (SERS) is a highly sensitive and high-resolution molecular recognition technique with important applications in many fields. As an emerging low-cost, high-resolution, and highflexibility micro-nano processing method, femtosecond laser direct writing has been widely used in the field of preparing SERS substrates. Compared with traditional processing methods for preparing SERS substrates, femtosecond laser direct writing processing has certain advantages in terms of flexibility, three-dimensional molding, processing material range, processing accuracy, and other aspects. In this review, we classify the processing methods of femtosecond laser preparation of SERS substrates into four categories, including femtosecond laser two-photon metal reduction, femtosecond laser cutting metal, femtosecond laser cutting-sputtering, and femtosecond laser 3D printing. Femtosecond laser two-photon metal reduction uses the two-photon reduction effect to reduce metal cations in metal solutions to metals, such as silver ions in silver nitrate solutions to silver nanoparticles. This method is suitable for the one-step preparation of SERS substrates in closed microchannels. Femtosecond laser cutting metal directly prepares the SERS substrate structure on a metal substrate. This method takes advantage of the high peak power of the femtosecond laser to ablate the surface of the metal sample to obtain a patterned surface structure. At the same time, femtosecond laser ablation produces particle fragments, which are usually redeposited on the patterned surface, resulting in SERS "hot spots". Femtosecond laser direct cutting of metal can prepare SERS substrates in one step, which has the advantages of high processing efficiency and simple processing and is more conducive to the application of large-scale production of practical SERS detection. Femtosecond laser cutting-sputtering is to process any structure on non-metallic substrates such as polymers and then sputtering/evaporating metal nanoparticles on the surface of the structure. This method can prepare transparent and flexible SERS substrates, which are rich in application scenarios. Femtosecond laser 3D printing is to use the three-dimensional processing ability of femtosecond lasers to obtain rich "hot spots" by designing the structure of SERS substrates, and then using template-guided self-assembly technology with different driving forces to deposit/evaporate metal nanoparticles at designated locations. In this paper, we first introduce the current methods for preparing SERS and then conduct a comprehensive review of the processing methods of four femtosecond lasers to prepare SERS substrates. Finally, the advantages and disadvantages of the four femtosecond laser preparation methods for SERS substrate are briefly summarized, and the development prospects of this technology are prospected, aiming to provide it for future related research.
Abstract
Surface-enhanced Raman spectroscopy (SERS) technique plays an important role in molecular recognition fields due to its highly sensitive and high-resolution. As an emerging low-cost, high machining accuracy, and high-flexibility processing method, femtosecond laser direct writing processing has been widely used in the field of preparing SERS substrates. This work introduces four methods of preparing SERS substrates by femtosecond laser direct writing, including femtosecond two-photon reduction, femtosecond laser cutting metal, femtosecond laser cutting-sputtering, and femtosecond laser 3D printing. The article introduces the performance and application scenarios of each method in preparing SERS substrates and illustrates the advantages of femtosecond laser direct writing processing in preparing SERS substrates, aiming to provide a reference for future related research.

1 引言

在过去的几十年中,体外器官培养、精准医疗、环境监测、化工、材料分析等领域发展迅速,因此需要快速准确地鉴定分析物的组成成分及含量。传统方法有紫外分光光度计、光致发光、荧光法、红外检测等检测方法,但其一般需要特定的仪器设备,且多是宏观、大区域检测,难以同时满足局域、定点、微量的传感需求。表面增强拉曼光谱(Surface-enhanced Raman spectroscopy,SERS)在实现非破坏性检测、制备样品简单、超高灵敏度(低至单分子指纹信息[1-3] )等方面具有独特的优势。表面拉曼光谱通过在基底表面形成不同的纳米结构(纳米间隙[4-7]、纳米星[8-10]、纳米棒[11-13]、纳米颗粒[14-17]等),在不同环境下(如环境检测[18]、食品检测[19]、细菌检测[20-21]、生化分析[22-23]等)有着广泛的应用前景。

飞秒激光直写(Femtosecond-laser direct writing,FsLDW) 作为一种有前景的三维微纳加工技术,目前已经成为多种材料可设计性微纳结构的纳米加工技术[24-29]。飞秒激光直写技术具有无掩模加工能力、任意形状可设计性、高空间分辨率、易于集成以及适用于各种非平面衬底等特点。在过去的十年里,在FsLDW技术的帮助下,金属微/纳米结构的制备和集成取得了巨大的成功,是目前制备微纳复合多级结构有效的方法之一[24]。相比于化学合成和其他成型方法,飞秒激光加工SERS基底结构更加均匀稳定;与反应离子刻蚀、电子束光刻和金属蒸镀等制备方法相比,飞秒激光的加工工艺步骤少、成本更低、效率更高。飞秒激光制备SERS基底最大的优势是能够制备丰富的可设计的微纳米结构,利用飞秒激光三维灵活的特点,能够在特定的位置产生丰富的“热点”区域,因此飞秒激光在制备SERS基底方面是比较有前途的方法之一。

本文简单介绍了飞秒激光微纳加工在SERS基底制备中的加工方法,并且对不同飞秒激光加工方法进行了对比,分析了飞秒激光对金属微/纳米结构功能器件的贡献。将飞秒激光制备SERS基底领域的应用大致分为以下四类(图1):飞秒激光双光子金属还原制备SERS基底、飞秒激光切割金属制备SERS基底、飞秒激光切割-溅射制备SERS基底、飞秒激光3D打印制备SERS基底。文章对飞秒激光制备SERS基底领域的四种方法进行对比、分析,总结了各自方法的优点与不足,旨在为今后的相关研究提供帮助。

2 SERS研究现状简介

2.1 SERS技术简介

表面增强拉曼光谱(SERS)是一种应用广泛的传感技术,当分子被吸附到波纹金属表面(如银或金纳米颗粒)时,分子的非弹性光散射(图2(a))大大增强[33]。1974年Fleishmann等人发现当分子被吸附在有波纹的金属表面(如金或银)上时,分子的拉曼散射被大大增强,他们将这种增强归因于表面积效应[34]。1977年Jeanmaire和Van Duyne[35]等人对这一现象进行深入研究并解释了该现象的增强机理,将此现象称之为表面增强拉曼散射效应,并将得到的拉曼光谱称为表面增强拉曼光谱(surface enhanced Raman spectroscopy,SERS)。

在40多年的发展过程中,SERS的理论基础取得了很大的进展。当分子非常接近某些SERS活性纳米结构时,分子的拉曼信号会大大增强[37]。SERS基底通常采用由Ag、Au、Cu等金属组成的等离子体结构,其中金属导带电子在光激发时会集体振荡产生局部表面等离子体共振(localized surface plasmon resonances,LSPRs)。LSPRs与入射光线的耦合导致了质子纳米结构周围的二次电场,产生了质子热点,有效地将金属表面(距离为10 nm)的电磁场集中102~105。这种现象也被称为SERS电磁增强(electromagnetic enhancement mechanism,EM)机制,是SERS信号在103~108范围内增强的主要贡献者[36](图2(b))。除了电磁增强,SERS还可以通过化学机制(chemical mechanism,CHEM)增强达 103。CHEM产生于分析物和金属表面之间增强的偏振性(以及因此产生的SERS信号)[38],这种增强机制在很大程度上取决于分析物的化学性质和分析物与表面的相互作用/亲和力。因此CHEM的适用性被限制在较窄的化学物种范围内,如硫酸盐分子等。在SERS测量中增强因子(enhancement factor,EF)与分析增强因子(analytical enhancement factor,AEF)是量化整体SERS增强的两种重要指标[39-40]。EF通过比较SERS和正常拉曼散射(normal raman scattering,NRS)信号强度(I)与单位面积内吸附的目标分子数量(N)来量化信号增强(如式(1))。

AEF则是从比较SERS和正常拉曼散射(normal Raman scattering,NRS)信号强度(I)与分析物浓度(C)量化信号增强(如式(2))。

大多数情况下化学增强机理的增强因子为 103之间[41],电磁在增强机理的增强因子可以高达 106~ 1012之间[42]。因此SERS增强机理中电磁增强一般起到主导作用,而其增强效果与SERS基底表面结构的形貌密切相关,特别是其纳米结构的尺寸与形状,很大程度上决定了SERS增强因子的大小[43]。等离子纳米结构材料附近的EM场通常是非均匀分布的,SERS热点大部分产生在高度局限于空间的狭窄区域中,如纳米尖端、颗粒间纳米间隙或颗粒-基底纳米间隙[38]等,小部分SERS热点在纳米颗粒结点和支持等离子体共振的平坦金属表面产生,因此其场强大小取决于间隙距离和其他几何细节。电磁场的振幅与间隙大小反相关,而 SERS热点的延伸范围在2~10 nm之间。当把纳米颗粒间隙距离相关到1 nm以下时,非局域效应开始发挥作用。此外,在1 nm以下的小间隔中,由于电磁场增强过大,可能导致光学响应成为非线性(即非线性效应的阈值相应地减少,与场增强成反比)。在这种强耦合制度下,分子-纳米粒子系统的内在属性可能会发生改变,这反过来又会影响到SERS的强度[33]。因此在设计SERS衬底过程中,对亚纳米到纳米大小热点中的拉曼过程与竞争过程进行准确具体的建模是十分重要的。

2.2 当前SERS基底制备方法

目前SERS微结构加工方法主要可以分为自上而下微加工和自下而上自组装两类。

在自上而下微加工制备 SERS 结构方面,国内外多个研究组开展了相关研究[44]。复旦大学杨新菊教授和美国德州大学奥斯汀分校郑跃兵教授合作利用反应离子刻蚀(reactive ion etching,RIE)加工硅纳米柱粘附金纳米颗粒制备了大面积的 SERS 结构,实现了高重复性和空间均匀度的超灵敏分子检测(图3(a))[45]。国家纳米科学中心褚卫国教授等人利用电子束光刻(electron beam lithography,EBL)、化学气相沉积和金属蒸镀的方法制备了多种几何结构的纳米 SERS 单元,通过将三维周期金/二氧化硅纳米栅模型的理论和实验相结合,提出了一种高性能三维表面增强拉曼散射探头的通用设计方法并实现了高精度的 SERS 检测(图3(b))[46]。南京师范大学吴平团队利用EBL制备了热点可控且表面开放的金纳米孔阵列,通过将有限差分时域模拟与实验相结合,提出了一种能够测定DNA甲基变化的SERS平台(图3(c))[47]。自上而下微加工方法可设计性强,且能够制备出三维SERS基底,但通常需要依赖昂贵的设备和复杂的加工步骤,因而成本较高,且加工精度存在极限。

因此,自下而上的加工方法得到了研究人员的重视。纳米颗粒自组装作为一种典型的自下而上加工方法,其在制备 SERS 基底中的应用得到了广泛关注和发展[51-53]。2010 年 Fan 等人在 Science发表论文,提出用核壳结构的纳米颗粒自组装制备具有等离子增强功能的团簇结构[50]。美国 USC 的研究人员引导金纳米颗粒 (Au nanoparticles,AuNP)组装形成 SERS 热点用于细胞成像研究,提供了一种在生物标靶上远程组装纳米团簇探针方法,可用于细胞内和体内的高灵敏度和高选择性的多模式SERS和光声成像[48]。合肥工业大学刘洪林教授使用油-水界面引导纳米颗粒排布形成 SERS 热点,用于食品农药残留的检测研究[49](图3(d)~3(f))。此外,研究人员还将纳米颗粒形成的 SERS 热点与微流体技术结合,开发了可用于实时分析的微流体 SERS 技术[54]。利用纳米颗粒自组装制备 SERS 基底的工艺流程简单,但存在样品污染、结构可控性差等问题。在溶液环境中测量时,难以避免布朗运动等干扰,信号稳定性差。

为了解决纳米颗粒自组装方法中的随机扰动等问题,研究人员结合自上而下微加工和自下而上自组装,提出了微柱自组装的复合加工方法[55]。此种方法是首先使用EBL、聚焦离子束(FIB)、纳米压印等手段加工出刚度较小的金属材料或聚合物/金属复合材料纳米柱结构,随后滴加液体;在液体蒸发时,如果纳米柱受到的毛细力大于纳米柱结构支撑力,则结构自组装形成 SERS 热点结构[21, 55-60](图4)。这些方法虽然分别在纳米间隙可控性、间隙介电材料选择等方面有一定突破,但其微/纳米柱的制备依然依赖于昂贵的 EBL 和 FIB 设备。此外,在 SERS 结构的实际应用中,往往需要在微通道等非平面基底上实现其加工和集成,但目前基于 EBL 和 FIB 的加工方法难以实现在非平面基底上的 SERS 结构制备。

目前,上述三种方法(自上而下、自下而上和微柱自组装)一般可以在一个或若干方面解决特定 SERS 功能结构的某些加工问题,实现高精度的 SERS 传感,但需要依赖昂贵的实验设备或繁多的工艺流程,且多数只能在平面基底上实现 SERS 结构,难以集成到微通道或光纤的特定位置。综上所述,非平面基底上 SERS 结构及器件的集成加工仍然是一个难题,这限制了 SERS 功能结构的进一步应用。

3 飞秒激光加工制备SERS结构方法

相比于上述的加工技术,激光加工是一种新兴制备SERS基底的技术[61-63],其具备灵活性高、无需模板、成本低等优点。而在激光加工方法中,飞秒激光加工有着独特的特点:1) 热响应区域小,加工精度高。当激光与材料相互作用时,能量从电子转移至晶格中,自由电子与晶格的热传递时间取决于材料中电子-声子耦合时间(1~100 ps)。当激光脉宽短于耦合时间时,大部分激光能量被电子吸收,迅速被转移给晶格而无热扩散损耗[64]。当飞秒激光作用在材料上时,由于飞秒激光的脉冲宽度非常短,能量在短时间传递给晶格,从而没有热扩散损耗,所以飞秒激光有非常低的热效应,因此飞秒激光具有加工质量高的特点。2) 加工材料范围广,几乎适用所有材料。由于飞秒激光的峰值功率极高,可达TW( 1012 W)量级,因此飞秒激光可加工多种材料,包括金属、聚合物、陶瓷等[25, 65]。3) 非线性多光子吸收。单光子吸收是当单个光子的能量超过材料带隙时,材料的电子吸收一个光子被激发到激发态,这是一个线性过程。但当光子密度极高时,即使单个光子的能量低于材料带隙,材料的电子也会同时吸收多个光子,从而达到激发态,此过程被称作多光子吸收,是一种非线性过程。材料非线性吸收仅发生在功率密度足够高的飞秒激光焦点附近,因此加工精度也得到了提高[66-67]

由于飞秒激光具有精度高、灵活性高、材料适用性广和无需真空环境等优点,因而得到了广泛应用。飞秒激光直写相比于其他方法[26, 68]具有三维加工能力、无掩模加工、适用于非平面衬底等独特优势。因此在SERS基底加工领域利用飞秒激光灵活三维集成的特点,能够设计加工出多种不同结构的SERS基底。目前飞秒激光制备SERS基底的加工方法主要分为以下四类:飞秒激光双光子金属还原、飞秒激光切割金属、飞秒激光切割-溅射、飞秒激光3D打印。

3.3 飞秒激光双光子金属还原制备SERS基底

飞秒激光双光子效应发生在当光强足够强大的情况下,材料同时吸收两个相同能量的光子或者在非常短的时间内连续吸收两个光子。与单光子吸收相比吸收相比,双光子效应仅发生在激光焦点位置附近,因此具有更高的分辨率[69]。利用飞秒激光双光子效应能够诱导金属盐溶液的光还原反应,例如将硅片上硝酸银溶液中的银离子还原成银微纳米结构,其原理(如图5(a))是飞秒激光对硅中价电子的强烈激发导致了等离子体羽流的绝热爆炸,由此产生了大量的活化物种(电子、离子、原子、团簇等)。在 AgNO3溶液中,银阳离子( Ag+)在硅等离子体和液体的界面上发生还原反应,通过在等离子体膨胀期间捕获上游电子,产生中性粒子( Ag0)。同时,激光诱导的硅等离子体可以激发水等离子体产生水合电子和自由基,进一步将 Ag+还原成银原子[70]

2010年孙洪波教授团队开发了一种灵活的飞秒激光写入路线,在非平面基板上排布金属纳米线图案化结构[71]。利用飞秒激光诱导银离子溶液还原,在非平面基板上加工出任意的电路或导线(如图5(b)),并且成功在微通道内的制造微加热器,展现了对微流体反应的理想加热能力,为MEMS (microelectromechanical systems)、LOC(lab on a chip)等智能微系统中集成微电路的设计和制造开辟新的途径。2022年华中科技大学甘棕松教授利用飞秒激光双光子还原制备了具有高均匀性的尺寸仅有230 nm的低电阻率铜纳米线[75]。因此双光子还原这一技术具有制备纳米电子器件的前景。2012年张永来教授等人提出了一种基于微流体通道的银微花阵列的片上催化微反应器和同步原位SERS监测器[72]。通过飞秒激光双光子还原诱导还原银溶液,在微流控通道中制备出直立的纳米板和附着的纳米颗粒构成的银微花阵列结构,观察到了4-NP还原为4-AP的化学反应过程(如图5(d)),且具有较高的SERS增强效应,可用于实时化学反应监测。

飞秒激光双光子诱导金属还原这一策略实现了在微流控通道中直接继承SERS基底,因此这项工作之后研究人员又开展了一系列工作。2017年吉林大学陈岐岱教授等人针对微通道中的银微纳米结构易氧化导致其稳定性与寿命下降的问题,利用飞秒激光诱导共还原的方法[73](如图5(e)),采用不同的前驱体溶液,在微通道中成功制备了不同含量比的银-钯(Ag-Pd)合金纳米结构,利用Ag-Pd结构形成合金保护SERS基底不易被氧化。在制成的具有18%钯含量的银钯合金基材保持了约2.62× 108的增强因子,且在日常环境下能够维持20天的使用寿命,这项工作为提高SERS基底的抗氧化能力提供了新思路。2017年首都师范大学李志鹏教授团队采用两步光还原法制备微流控单分子SERS设备(如图5(c))[74]。首先使用飞秒激光还原银离子,在微通道中制备了一个初级银纳米粒子聚合点,清洗通道后继续使用飞秒激光对聚集体进行第二次辐照。此方法解决了微流控通道中的SERS检测物易粘附在通道壁的问题并且将检测浓度下限提升到 1013。这种单分子微流控SERS设备能够降低对分析物的数量要求,且提高检测的可靠性,可用于快速和超痕量的SERS检测以及分析和诊断的实时监测。2018年日本理化研究所(RIKEN) Koji Sugioka教授提出了一种在3D玻璃微流控通道内使用飞秒激光加工制造2D周期性金属(Cu-Ag)纳米结构的新技术[76]。在微流控通道中飞秒激光直写选择性沉积铜银(Cu-Ag)层薄膜,再通过线性偏振激光束的照射,使微通道中形成由飞秒激光诱导的二维周期性金属纳米点阵列,得到一个具有灵敏度高、均匀性强的SERS基底。由此产生的SERS微芯片能够检测罗丹明6G,其增强因子为7.3× 108,并且还实现了在浓度10 ppb的镉溶液下进行SERS检测,这项技术能够制造高性能的微流控SERS平台,在实时感应环境中的有毒物质有很重要的应用前景。2019年英国赫瑞瓦特大学Mark MacKenzie团队报道了一种在熔融二氧化硅衬底上通过激光还原银溶液获得银纳米颗粒的制备方法[77]。飞秒激光在放有硝酸银溶液的熔融石英表面上多次扫描,使其表面上形成均匀且密集、直径约为50 nm的银纳米粒子,对罗丹明6G和CV的检测极限是 1011 mol,增强因子最高为1.4× 1011。此方法实现了在微流控通道中原位制备SERS表面,避免了之前方法需要预先将分析物与胶体混合的操作,在微流控通道表面排列着可控的银纳米颗粒,具有高灵敏度和较大的SERS增强效应。因此制备出的微流控SERS设备具有检测流动液体中的微量物质的前景,且有希望在定制的三维光流控或微流控设备中进行广泛的生物和分子传感或表征实验。

飞秒激光诱导金属还原不仅能够在微通道中制备SERS基底,同时也能够在光纤等特殊材料上制备SERS基底。2016年深圳大学李学金教授提出一种基于聚甲基丙烯酸甲酯(polymethylmethacrylate,PMMA)制备SERS基底的方法[78]。利用飞秒激光烧蚀被切开的光纤,在其粗糙表面利用双光子还原将银溶液还原成银离子分布在光纤结构上,以此得到了SERS光纤光谱探针。2018年李学金教授在此前的基础上完善新型锥端光纤SERS探头的研制和应用[79]。由于尖端光纤具有比较小的尺寸,相比于之前的光纤结构,新型的锥端光纤SERS探头具有较高的灵敏度,对于罗丹明6G溶液检测的浓度下限为 109 mol。柔和的光纤SERS探针在特定的生化分析中具有广泛的应用,因此SERS光纤探头为SERS信号的现场测量和进一步制作聚合物SERS微芯片提供了一种替代方法。

通过飞秒激光双光子还原制备SERS基底的灵敏度较高,检测下限大多数在~ 109 mol,其最大的优势是能够在封闭的微通道中一步加工制备SERS基底,但是由于在制备SERS基底的过程中引入具有流动性的液体,飞秒激光双光子还原法难以保障基底表面纳米微粒结构的均匀性与稳定性,因此在大规模生产方面受到限制。

3.4 飞秒激光切割金属制备SERS基底

飞秒激光烧蚀因无掩膜、无接触的优点得到了广泛应用[80]。当飞秒激光脉冲冲击金属表面时,由于逆轫致辐射机制脉冲能量首先会在厚度约为10 nm的皮肤层上吸收。大多数情况下,由于电子-电子相互作用时间很短,可以认为激发电子的热化是瞬时发生的。因此金属中非平衡系统可以看成是两次亚平衡系统,即热电子和冷晶格[81]。随着电子-声子相互作用和电子扩散出激发区,这种暂态双温系统会在几皮秒内达到平衡[81-83]。当晶格温度升高到足够高时,可能会发生熔化和烧蚀(膨胀),并且熔化仅发生在几皮秒到一百皮秒之间。材料烧蚀通常在飞秒激光脉冲后几十皮秒[84-85]开始,并可持续到几纳秒,产生的羽流由中性原子、离子、团簇和纳米颗粒组成[84-88]。烧蚀后样品表面以非常高的速度冷却,约为1013~1015 K/s[89],并迅速重新凝固,形成表面结构(图6(a))。并且飞秒激光烧蚀自然会产生颗粒碎片,这些碎片通常会重新沉积到图案化表面上[90]。在SERS 应用中,这种重新沉积的金或银纳米颗粒碎片能够对拉曼信号起到增强作用。

2011年台湾中兴大学蔡毓楨教授采用一步法制备粗糙Ag表面。利用飞秒激光诱导周期性表面随机纳米结构(纳米腔、纳米球),制备的SERS基底能够检测最低浓度为 108 mol的罗丹明6G溶液[91] (图6(b))。在加工过程中没有引入其他的Ag氧化物杂质,实现在水溶液中对R6G的原位测定,与其他形式的SERS活性衬底相比,其优点是加工过程无污染、程序简单及能够处理复杂的形状,有开发敏感光学化学传感器、纳米电子学和生物医学设备的应用前景。2018年西北工业大学张文定教授等人利用飞秒激光对金(Au)薄膜进行网格状扫描来制造金纳米(Au-NP)基底,以实现拉曼光谱增强[94]。飞秒激光扫描金薄膜的过程中,纳米粒子会直接沉积在金膜表面,这为SERS检测提供了丰富的“热点”。Au-NP基底对浓度低至 109 mol的罗丹明6G(R6G)分析物分子的拉曼光谱有明显的增强作用。此外,还测量了浓度低至 109 mol的罗丹明B(RB)和孔雀石绿(MG)分析物分子的拉曼光谱,这两种目标分析物的所有特征峰都被清晰地观察到,证明Au-NP基底具有良好的可靠性。

在污染物监测、爆炸物检测和疾病早期诊断中对超低浓度溶液中分子的检测是十分重要的。表面增强拉曼散射(SERS)结合超疏水表面能够突破扩散极限,实现飞/阿摩尔范围内的检测,但是传统的制造方法过程比较复杂且昂贵,而超快激光在加工材料表面有着独特的优势。2020年清华大学钟敏霖教授团队利用飞秒激光直写协同氧化和氟化法在金属铜表面直接制备了一个超亲水-超疏水图案化基底结构[30](图6(c))。该平台在蒸发过程中提供了稳定的液滴固定和持续的Cassie-Baxter状态,因此分析物富集且大多数分子沉积在所需的中心区域。作者还将目标分子与三种类型的胶体混合在一起(银纳米颗粒、金纳米颗粒和金纳米星),所有的胶体都表现出良好的增强效果。其中金纳米星显示出最低的检测极限浓度(对R6G分子的检测极限为 1018 mol/L)以及最高的增强因子(EF = 1.09 × 1014)。

针对目前Au/Ag SERS基底存在着稳定性不足的问题,其原因是金属在空气中易被氧化导致检测灵敏度下降,从而导致稳定性不足。2021年清华大学罗晓等人利用飞秒激光在氩气中直接切割金属制备SERS基底,通过在氩气保护下用飞秒激光加工银片,加工出的S-Ag-Ar结构在真空环境保存7个月后仍能与刚制备的S-Ag-Ar 基底的灵敏度保持相同[92](图6(d))。得到的微碗阵列被大量的纳米结构覆盖,其中纳米颗粒能够提供丰富的“热点”(图6(d)),其形成的原因是当飞秒激光焦点聚焦在加工金属材料表面时,在激光焦点处会产生极高的能量密度,此时金属会被气化形成等离子体结构飞溅至空中[95-96],当激光加工完成之后,处于等离子体状态的金属由于重力因素又会沉积微碗阵列当中,形成纳米颗粒[97]。由于其具有低成本、良好的灵敏度、均匀性、稳定性和普遍性等综合性能,在食品安全检测方面具有很大的潜力。

在生物医学方面,由于制造SERS基底常用的金、银金属的生物相容性不足,因此SERS在生物医学上受到了限制。Ti6Al4V(成分:6wt%Al,4wt%V,0.25wt%Fe,0.2wt%O(max),其余钛)是一种常用的α+β钛合金,具有高强度、低杨氏模数和在人体内的化学惰性,因此被广泛用作生物植入物[98]。2019年北京航天航空大学管迎春教授团队等人在生物医学级Ti6Al4V衬底上制备了一种分层的飞秒激光诱导周期表面(laser induced periodical surface structures,LIPSS)结构(图6(e)),可以通过SERS和表面增强荧光(surface enhanced fluorescence,SEF)提供双模(图6(e))增强光谱特性[93]。利用飞秒激光一步制备了具有微槽、纳米波纹和纳米颗粒组成的层次化LIPSS结构。由于表面等离子体激活剂和“热点”的共同作用,层次型LIPSS衬底表现出了超灵敏的检测能力,在 1011 mol/L浓度下,CV的增强因子为7.85× 105,在检测极限浓度为 1010 mol/L浓度下,层状LIPSS的LSPR和 "热点"使得基底上的CV荧光强度增强了约14倍。作者将SERS和SEF整合到单一的生物金属基底中,有希望提高生物医学调查中检测的灵敏度和可重复性。

目前飞秒激光直接烧蚀一步制备的SERS基底具有高效率、简单的加工优势,更有利于大规模生产和实际SERS检测的应用。但是飞秒激光直接烧蚀金属表面无法准确控制纳米粒子的形貌,其SERS基底表面的纳米间隙具有随机性,因此其在SERS基底的均匀性上有所不足。

3.5 飞秒激光切割-溅射制备SERS基底

切割-溅射制备SERS基底,首先一般在硅片[99-100]、石英玻璃[101-103]、铝片[104]等结构上加工出微纳米结构,再在其结构表面沉积金/银纳米颗粒或金/银薄膜使其具备SERS性能。利用飞秒激光独特的三维可设计加工特点,制备更加均匀、稳定的微纳米结构。2021年北京理工大学胡洁教授团队利用飞秒激光改变基底材料表面特性,简单有效地控制电镀(galvanic replacement,GR)反应中金(Au)和铂(Pt)的沉积点,制备出了大面积的硅基SERS基板[105](图7(a))。此方法能够简单控制金属纳米颗粒之间的间隙,与单独GR技术制备的金属纳米颗粒相比有着更高的SERS增强因子,最高为8.46× 107,并且具有良好的信号均匀性(相对标准差 < 9.7%)和化学稳定性(空气中2个月后强度偏差4.8%)。

除了制备大面积SERS基板之外,也可以利用飞秒激光诱导等离子体辅助烧蚀(LIPAA)制作透明柔性SERS基底。激光诱导等离子体辅助烧蚀(femtosecond laser induced plasma assisted ablation,LIPAA)是一种在透明基底上通过激光束聚焦在目标材料上以产生等离子体的方法[109],当激光能量高于目标的烧蚀阈值和低于基材的损伤阈值时,激光激发的等离子体主要在垂直于目标表面的方向上膨胀[110]。由于激光束和激光激发的等离子体与靶材表面的基材相互作用,放置在靶材前面的透明基材可以被烧蚀或标记[111-112]。2021年新加坡国立大学洪明辉课题组通过飞秒激光等离子体辅助烧蚀一步法制备柔性氟化乙烯丙烯表面等离子体共振膜的方法,用于原位SERS检测(图7(b))[31]。通过把柔性氟化乙烯丙烯(FEP)膜放置在带有间隔物的金属钯顶部,将飞秒激光聚焦到金属钯上导致金属材料蒸发,蒸发的金属材料与激光束相互作用导致等温等离子体的形成和膨胀,由于等离子的各向异性膨胀,导致金属纳米颗粒在FEP膜上的沉积[113]。作者研究了飞秒 LIPAA 产生的银和金纳米粒子的分布和尺寸,FEP 薄膜上的 Ag 和 Au 纳米颗粒的尺寸分布和密度可以通过有效调节激光能量密度来优化,所提出的 Ag NPs/FEP 和 Au NPs/FEP 基板显示出的增强因子分别为 5.6 × 107 和 2.4 × 106,并且在苹果原位测定福美双的检出限为0.1 mg/Kg,这在检测食品安全方面有着巨大的潜力。2022年洪明辉团队在之前的基础上,同样是针对食品安全方面,探索了硅纳米粒子在由 fs LIPAA 制造的用于 SERS 检测的玻璃基板上的沉积(图7(c))[106]。通过调节激光通量,可以很好地控制纳米粒子的尺寸分布,在衬底表面镀上一层银膜,可以为表面增强拉曼散射功能化提供高均匀性的热点。与传统的光滑银膜相比,利用R6G拉曼探针制备的银纳米颗粒/玻璃SERS衬底具有良好的均匀性,增强因子为2× 105,奶液中三聚氰胺和水中环丙沙星的检测极限均为1 ppm。

由于待分析溶液分子无法准确的进入到SERS基板中“热点”位置,因此会导致大部分测试样品被浪费,只有一部分测试分子对热点有贡献。因此一部分研究人员开始关注特殊图案化SERS基底中,如疏水/超疏水图案化SERS基底和超亲水/超疏水图案化SERS基底[114],当含有分析物的液滴在水接触角超过150°、滑动角小于10°的低粘性超疏水表面上蒸发时,咖啡环效应能够有效一致,从而导致分析物分子的富集[114-116]。2018年重庆大学刘安平团队利用激光辅助烧蚀在PTFE表面上制备SERS基底[107](图7(d))。利用飞秒激光处理使部分原始PTFE表面由疏水变为超疏水,之后再将银胶溶液滴在结构表面上蒸发,从而得到活性SERS基底。2020年清华大学钟敏霖教授团队利用飞秒激光在不锈钢上制造具有设计图案的超疏水/疏水混合表面,实现快速和高灵敏度的SERS检测[117]。依靠疏水图案和周围超疏水表面之间的结构和液滴附着力的差异,分析物分子可以被充分富集到预定的位置,小面积的沉积分子可以很容易被捕获,从而快速完成SERS检测,实现了对浓度为 106 mol浓度的罗丹明6G溶液的检测。但是由于疏水表面对液滴的粘附力较低、定位作用较差,因此导致分子沉积范围过大,这影响了SERS基底的灵敏度。相比之下,超亲水/超疏水结构表面更加有利于目前分子的聚集。2022年深圳科技大学杨灿等人利用飞秒激光制备了混合超疏水/亲水(SH/SHL)微孔平台(图7(e)),在平台上沉积一层银膜得到超疏水/亲水SERS基底 [108]。依靠在SHL图案中心制造的微孔,减少沉积物的面积,使目标分子在热点中心富集。对5 µL的R6G溶液的检测极限浓度低至 1017 mol,增强因子为5.19× 1013,在浓度为 1010 mol时,拉曼信号的RSD低至7.48%。因此该SERS基底在超痕分析中具有广阔的应用前景。

飞秒激光切割再沉积所制备出的SERS基底的检测下限较好,可以制备透明柔性基底,在一些特殊的环境下有很大的优势。但是其在加工时需要先在非金属基底上切割再沉积金/银纳米颗粒,因此工艺性与均匀性受到限制。

3.6 飞秒激光3D打印制备SERS基底

飞秒激光双光子加工技术可以视作一种特殊的3D打印,即光固化成型技术。目前将光抑制与光聚合相结合,相关研究人员已经将飞秒激光直写的加工分辨率突破至52 nm以下[118]。根据光的电磁学本质,光能够传入透明介质,在材料内部加工,从而实现真三维的空间结构成型[119-120]。此性质被利用在SERS基底制备上,从而能够在不同材料上制备出任意结构的SERS基底,实现SERS基底在特殊环境的应用。

3.6.2 双光子直写结合金属蒸镀

基于光学元件的SERS传感结合了SERS和光学元件的优势,能够实现灵活、坚固和小型化的光谱工具,甚至在各种恶劣的环境中对低浓度的分子分析物进行远程和高灵敏度检测。2015年首都师范大学冯胜飞教授设计了一个由抛物面镜和三维球形SERS体组成的SERS雷达构架(图8(a))[121]。激光被抛物面镜聚焦到SERS体的表面,SERS体表面由附着金纳米颗粒的微米级沟壑组成,抛物面镜收集拉曼散射光并将其反射回光纤。SERS雷达结构对乙醇中水晶紫的检测极限为 106 mol。此方法提供了一种在光纤小平面上制备SERS结构加工方法,但该装置的设计(有少量的微米宽的开口)在生物分析物传感方面存在着一些不足。2020年伦敦帝国理工学院的Jang Ah Kim提出了一种基于光纤表面增强拉曼光谱探针[122](图8(b) )。利用飞秒激光双光子聚合技术在光纤的表面加工3D微纳结构,再在其表面蒸镀金纳米层。在SERS测量中,激光被耦合到光纤中并激发光纤探针微纳米结构表面的信号热点,当与分析物相互作用时,SERS信号就可以产生并被光纤传感器收集。通过优化光纤尖端的表面增强拉曼散射阵列的设计、几何形状和排列,获得了3.7× 107 mol (罗丹明6G)的探测极限和的3× 103增强因子。此方法制备出的光纤SERS探针只需要2.5 ms(平面基板,10次平均)就能够快速检测细菌,并且光纤传感器能够用于非标记检测和原位表征方法来检测细菌,因此有望被集成到医疗内窥镜中,用于对发炎组织进行分析。

3.6.3 双光子直写引导毛细力自组装

毛细力是自然界中存在于非互溶体系界面上由于液体内弯面而产生的作用力,广泛存在于宏观、微观和介质领域。在微纳系统中,由于尺度效应的存在,毛细力(capillary force,Fc)的作用会被大大放大,当其超过维持结构刚度的支撑力(standing force,Fs),结构便会在毛细力的主导下倒塌或汇聚[123-124]。飞秒激光打印技术具有快速、简单、灵活度及可调节性极高的特点,将飞秒激光打印技术与毛细力辅助自组装(laser printing capillary-assisted self-assembly,LPCS)相结合的方法用于实现具有高度可设计性的、多层级的功能结构的灵活加工[125]

中国科学技术大学吴东教授团队利用飞秒激光三维加工灵活的特点在软基板上构建层次结构[126] (图9(a))。飞秒激光在处理过的PDMS上直写加工制备微柱阵列,利用毛细力引导微柱自组装(CFSA),在软基板上组装成各种随意设计的复杂层次结构模式。然后将机械调谐策略与CFSA相结合,实现了CFSA结构的可控变换,并且证明了这种可调谐的自组装结构可用于显示不同的图案和对尺寸变化的物体进行微捕获。此方法避免了对多步骤工艺和昂贵设备的严重依赖,具有简单性、可扩展性和高度灵活性。但是研究仅停留在对简单机械结构的探索,未开发出实际应用。

2020年吴东教授团队在此前的基础上提出了可切换的毛细力自组装方法(capillary-force self-assembly, CFSA)[7] (图9(b))。首先控制飞秒激光在光刻胶上加工出已设计好高径比的微柱阵列,在正常情况下,微柱阵列会由于毛细力而自组装,为了防止这种情况,作者通过采用超临界干燥的方法,使得聚合物材料在毛细力组装前可以保持直立状态,之后在聚合物微柱上涂上金纳米膜,这使得聚合物微柱成为聚合物-金属化合物结构样品浸入溶剂后,聚合物-金属化合物柱通过 CFSA 弯曲在一起,形成金属纳米间隙。此方法具有出色的灵活性,可以在非平面基底上制造各种图案化的等离子体纳米间隙结构。作者将此加工工艺运用在微通道中,其对罗丹明6G SERS信号的最大增强因子为 8×107,并且在微通道中实现了对抗癌药物(阿霉素)的局部传感。

毛细力自组装的方法能够在平面/非平面基底上设计出任意结构,通过巧妙运用毛细力自组装形成的间隙提供丰富的热点,并且在微流控芯片中的原位监测方面具有潜在的应用价值。但是由于加工步骤较多,不利于大规模生产,这限制了其在社会中的应用。

3.7 各方法对比分析

上述的四种飞秒激光制备SERS方法有着各自的优势,可以从加工维度、加工效率、SERS基底三个方面进行对比(表1)。从加工维度方面对比,双光子还原金属与毛细力自组装两种加工方法能够制备三维结构,因此在生化分析等方面具有广泛的应用场景。飞秒激光切割金属与切割-溅射方法大部分是制备二维SERS基底,通过在金属表面加工或其他材料表面加工再溅射金属的方法制备SERS基底。虽然是二维结构,但是其能够直接在表面制备不同形貌结构,以此获得纳米间隙、纳米尖端、纳米星等多种SERS增强符合结构,因此具有较低的检测浓度下限。从加工效率上比较,由于双光子金属还原与毛细力自组装加工方法是利用飞秒激光双光子还原的原理进行加工,其加工功率较小仅十毫瓦左右,加工精度达到纳米级,因此优势是能够得到微米级的SERS基底结构,并且能够在微通道中直接加工出设计结构,但是双光子还原仅仅发生在激光焦点处,因此其加工效率不高。飞秒激光直接切割金属的加工由于是飞秒激光直接在金属上制备SERS基底,因此激光的能量在百毫瓦级别,激光能量较大能够快速的制备大面积SERS基底结构,但是由于激光能量较大,其表面结构易受到热效应的影响,因此制备出的基底表面结构的均一性不足。从制备特殊基底方面比较,双光子还原能够在光纤、聚合物、开放微通道中制备SERS基底;飞秒激光切割金属则是在不同的金属上制备SERS基底;飞秒激光切割-溅射能够在柔性基底、刚性基底上制备SERS结构;毛细力自组装则是能够在聚合物、光纤端部、封闭微通道中制备SERS基底。

4 结论与展望

综上所述,本文简单分类介绍了制备SERS基底的方法,重点描述了飞秒激光制备SERS基底四种不同的加工方法,并进行了详细的介绍和对比。当前制备SERS基底加工方法大致可分为自上而下、自下而上、微柱自组装三类。自上而下微加工方法优点是可设计性强,可以制备任意结构的三维SERS基底,但是与激光加工技术相比,其加工步骤繁琐且加工设备昂贵,同时加工精度存在极限;自下而上加工方法优点是制备过程简单且能够结合微流控技术,但是其加工时存在着样品污染、结构可控性差等问题;微柱自组装加工方法在控制纳米间隙、间隙介电材料选择方面有所突破,但是其无法在非平面基底上制备SERS基底,同时也依靠昂贵的加工设备。飞秒激光加工在加工灵活性、三维成型、加工材料范围、加工精度等方面有着一定的优势,因此是一种新型加工SERS基底的方法。飞秒激光制备SERS基底可分为四类:飞秒激光双光子金属还原、飞秒激光切割金属、飞秒激光切割-溅射、飞秒激光3D打印。飞秒激光双光子还原金属与毛细力自组装加工方法是基于双光子还原基础制备SERS基底的,因此具有加工精度高、可加工三维结构、能够在微通道中加工等优势,但是此方法的加工效率较低,无法制备大面积SERS基底。飞秒激光切割金属与飞秒激光切割-溅射加工方法适用制备大面积二维SERS基底,通过直接加工金属表面可以得到不同形貌的热点结构,或者改变金属表面亲疏水等特性,使得分析溶液准确的进入到热点区,以此增强SERS效应,飞秒激光切割-溅射能够在柔性基底或者刚性基底上制备出SERS基底,在应用场景上比较丰富。

在未来的研究可以从以下几个方面进行突破:1) 针对双光子效率低的问题,利用空间光调技术提高其制备SERS基底效率;2) 利用飞秒激光制备三维光学器件加工能力,将光学器件在SERS结构上集成,提高检测效率;3) 继续发展新的SERS基底材料,提高其抗氧化能力与增强因子;4) 由于SERS在生物医学领域上具有广泛的应用前景,因此能够在生物体内获取拉曼信号的技术也至关重要。
