1 引言
计算机断层扫描(CT)成像技术以其无损检测的特性在生物医学[1-3]、新材料、微系统加工与制造、地质勘探、新能源、航空航天以及考古等诸多领域有非常广泛的应用,自问世以来受到了众多关注并获得了迅速发展。
1973年Hounsfield[4]搭建了第一台真正意义上的医学CT系统。此后较长一段时间,CT系统凭借其独特的无损检测特性获得了快速发展。从第一代的单射线源单探测器的平行束CT到现在的多探测器甚至多射线源的螺旋CT[5-6],扫描效率和扫描精度都得到了很大提高。受限于时间分辨率以及低剂量的要求,目前常规的医用CT最优分辨率仅有几百微米,然而这远不能满足高新材料、生物前沿以及微系统加工与制造领域对高分辨率的需求。20世纪80年代,类似临床CT成像结构的显微CT的空间分辨率达到了147 μm×147 μm×1000 μm。随着基于FDK算法[7]的重建技术的引入,显微CT的纵向分辨率得到了极大的提高。由于X射线具有高穿透性,难以进行调制,因此CT成像系统的分辨率的提高主要依赖于投影放大,这就要求射线源具备较小的焦斑尺寸,探测器有较小的像素尺寸。受加工制造技术的限制,X射线高分辨率成像的潜力得不到有效的开发,X射线CT的系统分辨率一直处于几十微米的水平。随着微米级焦斑尺寸的射线源和高分辨率的X射线探测器(如透镜耦合X射线探测器[8])相继研制成功,基于这两者构建的显微CT的最佳分辨率一般在百纳米量级,视场为毫米量级。为了缩小焦斑尺寸,澳大利亚CRSIO公司研制了基于扫描电子显微镜(SEM)的X射线源,实现了准纳米级焦斑尺寸。利用这种准纳米级焦斑尺寸的光源结合高倍率透镜耦合探测器搭建的Nano-CT成像系统可以实现优于50 nm的分辨率[9]。然而这种基于SEM的X射线源光通量极小,清晰成像需要的曝光时间很长,成像效率极低,并不适合长时间的CT扫描。目前市面上常见的商用纳米CT大都依赖微焦斑X射线源和高分辨率成像探测器,最优的分辨率能够达到200 nm。商用纳米CT通常采用具有微纳焦斑尺寸的实验室源作为光源,这种X射线源本身亮度低,导致高分辨率的CT扫描效率低,在一定程度上制约了X射线调制元件的研制与开发。
同步辐射装置的蓬勃发展极大地促进了X射线元件的研制与发展,使得X射线的光学成像特性得以有效开发。同步辐射X射线源具有高准直、宽波段、高亮度的特点,便于利用X射线元件进行调制。随着大型同步辐射装置的进步及各式各样的X射线聚焦元件如波带片和毛细管等的开发,对X射线的聚焦与放大得以实现,使得X射线成像分辨率提高到了纳米级别,从而实现了真正意义上的纳米CT成像技术[10-13],成像分辨率在10 nm量级,其成像视野(FOV)一般只有几十微米。目前世界上多数的同步辐射研究中心都配备了高端纳米CT成像装置,包括美国先进光源(ALS)、欧洲同步辐射中心(ESRF)和日本大型同步辐射光源Spring-8,国内的中国科学技术大学国家同步辐射实验室(NSRF)、中国科学院高能物理研究所(BSRF)、中国科学院上海同步辐射光源(SSRF)。纳米CT成像系统、技术研究和应用研发逐渐走向成熟。
2 纳米CT成像原理及分类
X射线与物质之间的相互作用是十分复杂的过程,在宏观上大致可以认为X射线在物质中发生折射,则有

式中:n为X射线对样品的折射率。吸收衬度与折射率的虚部β成正比,相位衬度与折射率的实部δ成正比。当X射线穿过样品后,不仅强度发生了变化,相位也同时发生变化,在像面产生吸收衬度和相位衬度,利用这两种不同的成像机制分别形成了基于吸收衬度的CT成像技术和基于相位衬度的CT成像技术。X射线波长极短,在0.001~10 nm之间,穿透力极强,因此透射型X射线吸收衬度显微镜发展快速,种类繁多,应用更加普遍。基于吸收衬度的CT成像系统按照放大方式的不同可以分为:基于X射线投影放大型的CT成像系统和基于X射线调制元件的CT成像系统,也就是X射线显微镜。
设I0为X射线的入射强度,I为X射线的出射强度。理想条件下,垂直入射的单能X射线穿过某均匀样品时满足
式中:Δx为X射线穿过样品的厚度;μ为衰减系数。 X射线穿透样品时与其内部的物质发生一系列相互作用的结果,经验上可以表示为μ(E,Z,ρ),其中E是X射线能量,Z是样品的等效原子序数,ρ是样品密度。由于样品材质和厚度不同,出射X射线的衰减量也不同,从而可以在探测器上获得包含样品内部结构的投影图像[14]。
CT成像系统的主要部件包括:X射线源、X射线探测器、调制元件、样品台及运动控制系统。扫描成像的过程是:通过X射线调制元件对射线进行聚焦后作为照明光源, X射线透过X射线调制元件进行投影放大,样品旋转360°,通过探测器采集到包含足够信息量的吸收投影图像或相衬投影图像,通过三维重建获得样品内部的三维信息。三维重建的过程可以简单理解为求取样品内部各离散点的衰减系数μ(E,Z,ρ)的过程。利用重建方法得到的某一个断层的二维截面,之后可以将所有的二维断层截面拼合起来,得到样品的三维图像。重建算法可以分为解析类重建算法和迭代类重建算法。解析类重建算法的重建速度较快,对于完整的数据能够得到高质量的重建图像,缺点在于要求扫描角度多、数据统计涨落小。迭代类重建算法适用于各种扫描模式,对扫描角度小、统计涨落大的数据重建得到的图像优于解析类算法,缺点是重建速度慢。典型的解析类重建算法有傅里叶变换重建算法、滤波反投影算法和反投影滤波重建算法;典型的迭代类重建算法有ART重建算法、Richardson重建算法等[14]。
纳米CT成像系统能够实现优于100 nm的分辨率,其成像系统结构精密复杂,有多种分类方式。按照成像机制的不同,纳米CT成像系统可分为基于吸收衬度的纳米CT成像系统和基于相位衬度的纳米CT成像系统;按照X射线性质的不同,纳米CT成像系统可以分为硬X射线CT成像系统和软X射线CT成像系统。依据系统结构的不同,纳米CT成像系统又可以分为基于投影放大型的CT成像系统和X射线显微镜型的纳米CT 成像系统。
基于吸收衬度的纳米CT成像系统在结构及重建算法方面与传统CT成像系统相似,成像装置结构简单,重建方法成熟,鲁棒性好,适用于对金属类、骨骼牙齿、芯片等原子序数较大的物质成像。然而,原子序数较小的生物软组织样本,对绝大多数波段范围的X射线的吸收能力相差不大,利用这种成像技术难以区分各种不同的轻质物质。
针对生物软组织如细胞等,基于吸收成像的水窗软X射线纳米CT成像有独特的优势。水窗软X射线是指能量位于284~543 eV的射线,在这个波段范围内,含碳的有机物对X射线的吸收是水对X射线的吸收量的10倍以上,对于大多数含水的有机物如细胞等,只需利用吸收成像便能达到很高的对比度。当然软X射线波长较大,景深小,成像范围受到严重限制,同时长时间的辐照对生物活体样品会造成损伤。
基于相衬效应的纳米CT成像技术是吸收型纳米CT成像技术的延伸和发展,对轻量元素组成的样品如生物软组织进行成像有独特的优势。这类样品所含元素的原子序数较小,在硬X射线波段内相移截面比吸收截面大2~3个数量级,因此利用相衬成像机制进行成像能获得更高的图像衬度以及更高的探测灵敏度。同时,硬X射线的相衬成像景深较大,约为几十微米,这样的景深可以完整成像大多数真核细胞[15-21],因而针对轻质材料,基于相衬成像机制的CT成像技术更适合。国家同步辐射实验室利用硬X射线显微术和纳米断层扫描在细胞成像方面的独特优势,开展了相关方面的研究,弥补了光学显微镜和电子显微镜在细胞成像方面的不足,这在国际上也是领先的探索,为将纳米CT成像技术发展成为一种有力的细胞成像工具做出了出色的工作[22]。
3 基于投影放大的纳米CT成像技术
基于投影放大的纳米CT成像技术主要通过几何投影放大直接进行成像,通过使用具有纳米焦斑尺寸的射线源和较大的几何放大比(源像距与源物距之比)达到纳米级别的分辨能力(
根据CT成像系统的有效射束宽度公式[14][(3)式],成像分辨率经验上为有效射束宽度的一半,由焦斑尺寸S、探测器的像元尺寸d和系统的几何放大比M共同决定:
射线源到待测样品的距离Z1越小,待测样品到探测器的距离Z2越大,则投影放大比M=(Z2 +Z1)/ Z1越大,越能够分辨更小的细节。焦斑尺寸S具有一定的尺寸,在投影过程中不可避免地会出现弥散斑,散斑尺寸为S',仅增加几何放大比并不能有效地提高分辨率。如何通过技术手段减小焦斑尺寸和像元尺寸成为纳米CT发展的重要方向,而这两者都依赖于X射线元件的研制。一方面,通过X射线元件能够实现对X射线的聚焦,从而实现焦斑尺寸的减小;另一方面,X射线聚焦元件能够像普通光学透镜一样实现光学放大,从而降低了对探测器像元尺寸的要求。
3.1 纳焦斑X射线源的获取
通常情况下,X射线源包括实验室X射线源和同步辐射X射线源两种。实验室X射线源亮度低,且焦斑尺寸越小,光通量越小,则成像效率越低。同时焦斑尺寸越小,X射线源的造价越昂贵,因此目前市面上鲜见具有纳米焦斑尺寸的商用X射线源。一种纳焦点射线源的实现方式是:将数十纳米的金属靶安置到改造扫描电子显微镜(SEM)源的真空腔内,电子束轰击金属靶,从而产生数十纳米焦斑尺寸的X射线源[23]。另一种是利用X射线元件聚焦实现纳焦斑,例如同步辐射X射线源配合X射线调制元件形成纳米焦点的射线源。
X射线聚焦元件按照原理可分为四大类:衍射型、反射型、折射型以及吸收型。

基于投影放大原理实现的纳米CT成像原理简单,但是对硬件的要求非常苛刻,极度依赖于具有纳米焦斑尺寸的X射线源。大部分X射线聚焦元件设计精密,加工困难,调制效率低,其研制与开发有赖于具有高通量的大型同步辐射装置。
3.2 高分辨率X射线探测器
高分辨率X射线探测器有光学透镜耦合型探测器和光纤型探测器两种,相较于普通的X射线平板探测器具有分辨率高的特点。光纤耦合CCD探测器结构如

3.3 样品台及运动控制平台
纳米CT采用的机械运动系统比显微CT的运动控制系统精度更高,但二者没有明显的界限。当前商用的线性运动位移台和旋转台的精度在数十到几百纳米不等[31],通常人们会采用各种外部方法检测运动误差并对其进行校准的方法来确保运动精度。2006年,Nicola等[32]利用电容式位移传感器在气浮转台上对转台的转动误差进行了校准,将转台误差降低到10 nm以下。Kim等[33]在2013年利用激光干涉仪作为距离传感器成功将转台的径向误差校准到40 nm以内。2014年Xu等[34]利用5个电容式位移传感器和一个标准件,通过重复实验,成功测出了转台转动时的系统误差,包括端跳、径跳、偏摆等。2017年Stankevi

4.1 X射线波带片
波带片被广泛用作软X射线波段的聚焦元件[24,38-45,57,73-80],利用波带片搭建的纳米CT成像系统如
波带片具有较高的聚焦效率,是典型的X射线调制元件,各研究小组开展了诸多工作以研究扩大其适用范围的方法。瑞士保罗谢勒研究所利用基板双面叠加技术加工制造了外径宽度为30 nm的波带片,利用能量为9 keV的硬X射线波段实现了30 nm的分辨率,效率达到了9.9%[24] 。Li等[40]结合金属辅助化学蚀刻和原子层沉积等最新技术在15 μm厚的硅晶片上制造出了厚度为8 μm、最小带宽为16 nm的铂波带片,宽高比达500∶1,对于20 keV的X射线的衍射效率为4.8%。这个数值比大多数菲涅耳波带板更高,对于如此高能量的X射线,其效率值接近用多层镀膜Kirkpatrick-Baez反射镜和多层劳厄透镜实现的效率值。美国阿贡(Argonne)国家实验室提出了基于平面镀膜制作技术的X射线多层膜劳厄透镜(multilayer Laue lens),其结构如

4.2 复合折射透镜
复合折射透镜是另一种常用的X射线聚焦元件,由多个特定形状的X射线凹透镜组合而成,主要分为球形复合透镜[

目前应用最成熟的是抛物面复合折射透镜,其适用能量区间广、安装简易,成为同步辐射实验站聚焦元件的首选。受复合折射透镜单位透镜表面的缺陷和堆叠精度的限制,这类X射线元件存在严重的球差,是X射线成像领域内的一大研究热点。越来越多的研究人员试图从光学的角度来理解和描述X射线,Seiboth等[88]对X射线透镜的波前误差进行了测定,并开展了校正工作,希望通过X射线的光学函数来评价和分析X射线成像过程,进一步了解X射线特性[89-90],从而能够从理论上对X射线断层成像提出进一步的发展方向。
4.3 布拉格放大器

5 基于相位衬度的纳米CT成像技术
基于相位衬度的纳米CT成像技术是吸收型纳米CT成像技术的延伸和发展,对轻量元素组成的样品如生物软组织进行成像有独特的优势。轻量元素组成的样品所含元素的原子序数较小,在硬X射线波段内相移截面比吸收截面大2~3个数量级,因此利用相衬成像机制进行成像能获得更高的图像衬度以及更高的探测灵敏度,这类成像技术比较常见的有泽尼克相衬成像、光栅相衬成像及Ptychofraphic(简称PTY)成像技术。其中泽尼克相衬成像和PTY成像技术对X射线的空间相干性要求极高,这对其发展造成了一定的局限性。另外一个非常关键的问题是相位信息的解析重构,目前主流的做法是引入微分相位衬度成像机制,以解决部分相位衬度定量化和相位周期性问题。相衬成像技术的成像机制决定了其抗干扰能力差,对系统装配精度和环境的要求很高。
X射线泽尼克相衬成像技术是泽尼克相衬显微镜在X射线波段的延伸,泽尼克相移环置于物镜波带片后焦面,使样品直通光产生π/2或3π/2的相移,进而与样品衍射光在探测器上发生干涉,将相位信息转换为强度信息被探测器探测到。同时这种成像技术有很高的灵敏度,是一种高分辨率、高衬度的成像方法[16,18,105-106],在细胞、生物组织以及有机材料成像中有着广泛的应用,成为生命科学、材料科学、信息科学、智能芯片、能源科学和纳米科技等热门研究领域不可或缺的研究手段[21,107-117]。
2003年Momose等[118]将X射线泰保-劳(Talbot-Lau)光栅干涉仪引入硬X射线CT成像系统,使X射线相位衬度成像技术的光源从同步辐射光源扩展到了常规光源。基于投影条纹的X射线光栅相衬成像方法对相干性要求低,光栅周期变大后可以降低光栅的制造难度,有望获得更大的成像视场以及更高的成像能量,在临床医学和工业扫描中难以避免受到震动和系统部件安装误差的影响,Hauke等[119]提出了一种增强重建算法来调整光栅的位置,从而补偿这些误差。
PTY成像技术在国内被翻译为层叠成像[120],成像原理是通过改变照明光束和样品的相对位置,获得系列交叠区域的衍射图样,恢复得到样品的振幅分布。X射线PTY既能充分发挥X射线的高穿透力,又能实现无透镜成像的高敏感度相衬成像[120-123]。生物组织样本、聚合物或纤维复合材料等轻质样品对X射线的吸收较弱,采用相位敏感成像的方法能够有效提高衬度,而常规的X射线吸收成像方法并不适用。层叠成像技术在大幅面成像和高分辨率成像方面具有巨大潜力。与传统的衍射成像相比,此成像系统无需透镜,不但避免了透镜的像差对成像恢复质量的影响,而且也消除了数值孔径对样品尺寸的限制。该技术不仅可以与多切片技术结合,消除样品的厚度效应,还可以与X射线荧光结合。但目前该技术依然存在着一些问题,比如样品太厚导致投影近似值的不确定性,最突出的问题在于其成像效率与质量之间的矛盾。
相比于传统的X射线吸收机制的成像技术,相衬成像技术仅经历了短短30年的发展,在成像装置和成像算法方面距离发展成熟尚有一段距离。然而,相衬成像技术对轻质材料内部成像有巨大优势,成为极有潜力的发展方向之一。
6 纳米CT成像现存问题
6.1 纳米CT向计量型的转化问题
对于具有内部特征的样品的尺寸测量,CT是少数适用的测量工具,随着CT成像系统相关标准的发展,CT已逐渐发展为计量工具。虽然目前纳米CT的最优分辨率已经突破10 nm,但是仅限于定性测量,还不能实现定量测量的功能。生物医学、高新材料、先进制造的快速发展对CT成像提出了从定性测量到定量测量的要求,成为纳米CT的主流发展方向之一[124-128]。然而,作为测量仪器,CT设备的校准规范及流程目前在国际上尚未达成共识,德国、美国、中国等国家发布了各自的标准,但并未形成ISO标准;针对CT测量的发展,德国Zeiss公司、Werth公司及日本的Nikon公司分别研制了部分计量型CT产品,但分辨率仅在数微米,纳米CT的计量功能的实现还需要大量的研究。
6.2 系统几何参数和机械定位精度
CT成像的重构过程是建立在理想几何位姿的基础上,然而实际上难以避免系统随机震动和装配误差给纳米CT的实现造成困难。对于利用同步辐射X射线源的纳米CT,X射线的获取和调制所需要的部件繁杂,进一步给调试和校正工作带来困难。采用实验室源的纳米CT系统则难以避免射线源的震动。除此以外,扫描过程中运动系统的震动会造成随机干扰。受机械加工水平的限制,旋转扫描模式还会造成扫描中心的偏摆。通常纳米CT相较于显微CT时间更长,对系统稳定性和系统部件的几何位置的精准度比显微CT有更加严苛的要求,然而目前对这方面的研究资料却不多见。
6.3 环境温度对成像的影响
纳米CT成像设备属于封闭式空间设备,在样品扫描过程中,射线源连续发射X射线,会引起内部环境温度的变化,热膨胀的存在使部件位置关系发生变化,造成图像分辨率下降。因此,在纳米CT成像设备中,需增加控温设备或对各部件位置进行实时监控。且对于采用实验室源的纳米CT,由于大多数纳米X射线源光强较小,扫描时间大大延长,普通探测器不可避免地会出现噪声增强的现象[129],对探测器暗电流的检测和校正对于提高系统的分辨率不容忽视[130]。
7 结束语
纳米CT成像系统在生物医学、微系统制造、材料学等多个研究领域已经成为与扫描电子显微镜、扫描隧道显微镜等一样不可或缺的成像仪器,目前已经能够实现亚细胞层级的三维成像。是最重要的无损检测手段之一。依托于同步辐射技术的进步,纳米CT成像技术获得了快速发展。世界上绝大部分的同步辐射装置均设置有CT成像系统,国际上的大型同步辐射装置主要分布在北美(ALS、APS、SLAC、CHESS、CLS等)、欧洲(ESRF、BESSY、SLS等)、东南亚(日本Springer-8、新加坡SSLS、韩国)及澳大利亚(AS)等地区。目前国内BSRF、SSRF、国家同步辐射实验室也在纳米CT成像领域做出了突出的贡献。研究人员基于SSRF在X射线聚焦元器件的制造领域进行了深入研究。国家同步辐射实验室建有中国第一台以真空紫外和软X射线为主的专用同步辐射光源。基于软X射线的纳米CT在纳米材料、细胞科学、多孔电极材料等领域中的应用研究取得了很多进展,填补了国内相关领域的空白[131]。理论方面,朱佩平等[132]在以吸收成像为机制的传统纳米CT中,引入折射和散射光强响应成像机制,提出具有三种定量成像机制的纳米CT三维成像方法,实现对直径5 μm以上细胞的完整三维成像,分辨率达到纳米量级。
纳米CT成像技术分辨率的提升依赖于X射线元件的发展。突破制造水平对促进纳米CT的研究和发展有重要的意义。同时,这种严苛的加工制造要求带来的高昂的经济成本也是纳米CT成像技术面临的困难之一。
X射线归根结底是一种光波,基于可见光光学成像的原理也可推广到X射线成像,得益于同步辐射装置和各种X射线元件的开发,纳米CT技术获得了快速发展,克服了光学显微镜的分辨率低和电子显微镜穿透性差的局限性,然而目前数十纳米的分辨率远未达到X射线的分辨率极限,其分辨率和稳定性还有较大的提升空间。
纳米CT是一种极其实用且拥有巨大应用前景的无损检测手段,能够实现定量检测是众多应用领域的需求,也是开发仪器的科研人员必然的努力方向之一。目前纳米CT领域的研究热点在于进一步提高系统的分辨率和提高系统测量的准确度。前者依赖于X射线成像元件的开发,后者则要求将成像系统的不稳定性降至足够小的范围。提高测量准确度的手段有许多工作可以开展,例如监测射线源的不稳定性曲线、机械系统的振动和噪声曲线等,对投影数据的调整有利于提高重建图像的质量,探究X射线与样品相互作用的热效应对尺寸测量的影响,这些工作的开展均有助于获取更加准确的图像,从而进一步实现溯源与计量。
