Coptidis Rhizoma (Chinese: Huanglian) and Phellodendri Chinensis Cortex (Chinese: Huangbo) are widely used Traditional Chinese Medicine, and often used in combination because of their similar pharmacological effects in clinical practice. However, the quality control methods of the two drugs are different and complicated, which is time consuming and laborious in practical application. In this paper, rapid and simultaneous determination of moisture and berberine in Coptidis Rhizoma (CR) and Phellodendri Chinensis Cortex (PC) was realized by near-infrared spectroscopy (NIRs) combined with global models. Competitive adaptive reweighted sampling (CARS) and successive projection algorithm (SPA) method were applied for variable selection. Principal component analysis (PCA) and partial least squares regression method (PLSR) were applied for qualitative and quantitative analysis, respectively. The characteristic variables of berberine showed similarity and consistency in distribution, providing basis for the global models. For moisture content, the global model had relative standard error of prediction set (RSEP) value of 3.04% and 2.53% for CR and PC, respectively. For berberine content, the global model had RSEP value of 5.41% and 3.97% for CR and PC, respectively. These results indicated the global models based on CARS-PLS method achieved satisfactory prediction for moisture and berberine content, improving the determination e±ciency. Furthermore, the greater range and larger number of samples enhanced the reliance of the global model. The NIRs combined with global models could be a powerful tool for quality control of CR and PC.

黄连素(BBR)在医学领域有广泛的应用, 其定量分析对指导临床治疗具有重要价值。 常见的黄连素检测方法的灵敏度、 响应范围、 检测成本等方面尚存在不足。 本研究发现, 带有负电基团的血卟啉单甲醚(HMME)对红区发射的强荧光化合物阳离子铝酞菁(TTMAAlPc)具有高效荧光猝灭作用, 形成几乎无荧光的离子缔合物, 而在盐酸小檗碱的存在下, 体系重新发出酞菁特有的红色荧光, 且荧光恢复程度与盐酸小檗碱的浓度呈正相关。 这一现象的发生是由于血卟啉单甲醚与阳离子铝酞菁分子通过平面疏水作用、 静电作用和水分子介导的氢键作用等超分子作用力而形成缔合物(HMME-TTMAAlPc), 而盐酸小檗碱则通过强竞争结合作用, 使阳离子铝酞菁从缔合物中被释放出来, 导致体系荧光显著恢复。 据此发现, 建立了盐酸小檗碱荧光增强定量分析新方法。 考察了各种因素对方法的影响, 在最优条件下, 标准工作曲线的方程为ΔIf=12.85c+66.30, 线性区间为0.3~69.0 mg·L-1, 检测限为15.0 μg·L-1, 相关系数r=0.997 2。 研究表明, 该方法特异性强、 操作简便、 成本低廉。 该方法已用于实际样品的测定, 取得了满意的结果。 该研究开拓了酞菁红色荧光探针在药物分析领域中的应用。

The content of berberine hydrochloride (BH) in compound berberine tablets (CBTs) is subject to strict requirements. Its content is usually measured based on chemical analysis. In this paper, the fluorescence spectral imaging method was used to study the relative content of BH from a physics perspective. By comparing the relative fluorescence intensity of self-made CBTs with different mass percentages of BH, a linear positive relationship was observed between the BH content and the relative fluorescence intensity, and accordingly the quality of CBTs of different brands was evaluated. The results indicate that the fluorescence spectral imaging method can be a simple, fast and nondestructive semi-quantitative analysis method to determine the content of BH in CBTs, and this method has great potential in the quality control of CBTs.

在pH 7.4的Tris-HCl介质中, 以盐酸小檗碱(BR)为荧光探针, 研究了紫草素(SHI)与鲱鱼精DNA的相互作用。结果表明, SHI与BR竞争结合DNA上的位点; SHI对DNA-BR体系的荧光有猝灭作用, 猝灭机制为静态猝灭和动态猝灭共存, SHI与DNA之间为嵌插和静电两种作用方式; 热变性实验进一步证明嵌插结合是SHI与DNA的主要结合模式。最后由Scatchard方程求得二者的结合常数为3.48×104 L·mol-1。

以盐酸小檗碱(BR)为荧光探针, 研究了鲱鱼精DNA与橙皮素的相互作用。采用荧光光谱, 紫外可见吸收光谱, 盐效应, Scatchard方程等手段, 探讨了橙皮素与DNA的作用机制. 结果表明, 在生理酸度(pH=7.4)下, 橙皮素对DNA-BR体系的荧光有猝灭作用, 猝灭类型为静态猝灭与动态猝灭共存的模式, 其作用方式为嵌插和静电作用的混合模式, 并求得其结合常数为1.83×104 L·mol-1.

中药提取物是传统中药融合现代制药技术的新型产品,具有广泛的国际市场。建立快速、有效的分析方法有助于中药提取物的实时质量监控。文章利用傅里叶变换红外光谱技术(FT-IR)对黄连药材、水提物和醇提物作了研究。结果表明:黄连药材及其提取物有着各自稳定的光谱特征,谱图中位于1 505 cm-1 的芳环骨架振动峰是判断盐酸小檗碱在不同黄连样品中含量高低的主要依据,而相应的二阶导数谱图可以大大提高原始谱图的分辨率。黄连药材经过提取,以小檗碱为代表的主要活性成分得到有效富集,两种提取物中小檗碱的含量明显增高;黄连醇提物中小檗碱的含量又明显高于其水提物。FTIR能够简便、快速地提供中药提取物中主要化学成分的定性与半定量信息,从而为后续的化学成分分析和中药提取分离工艺的改进提供有效参考。
