1 引言
神经科学的相关研究极大地推进了对神经系统控制行为这一机制的探索和认知,也促进了各类神经疾病发病机理的研究以及预防、治疗手段的开发[1-4]。对特定神经元进行高时空精度的调制和记录为上述研究提供了实验手段和数据支持。为了在相关领域取得突破性进展,科学技术部于2021年推出科技创新2030“脑科学与类脑研究”重大科技项目计划[5]。该计划的目标是通过对神经元进行高时空分辨率的调控和记录,以绘制高精度脑神经网络图谱,为神经疾病的预防和诊治、脑认知功能的基础原理等研究提供坚实的基础。为了实现这些目标,相应的神经元调制和记录技术也成为了神经科学领域的研究重点。
随着科学技术的发展,越来越多的方法被应用于对神经元的调制当中。常用的方法包括电刺激、化学刺激和光遗传刺激等。电刺激法通过可植入电极,在特定区域内刺激神经元,但是电刺激缺乏特异性,无法精确刺激特定的细胞,在治疗如帕金森症等疾病方面可能有致人抑郁的副作用[6-8]。而且,由于电流流经组织会产生大量焦耳热,在如人造耳蜗这样空间有限的应用场景下难以提升电极数量,不利于提高使用者对声音频率的分辨能力[9-11]。化学刺激能够通过药物和基因编辑等方法刺激、抑制特定的神经元,但是化学刺激达到效果的时间周期也会更长,并且也无法实现高时空精度的神经刺激[12]。磁刺激[13-14]和超声刺激[15-16]具有实现非侵入地调控神经元的能力,现已针对人体开展了较为广泛的应用与研究,但二者空间分辨精度有限[17]。而对于拥有复杂神经系统的脊椎动物,其神经元在生理行为、基因表达和神经连接等方面都有着很强的特异性。仅通过电刺激、化学刺激、磁刺激或超声刺激均无法做到单细胞精度的神经元刺激[2]。
光遗传刺激结合了光学和基因学手段,具有实现毫秒-亚毫秒量级的单个神经元调制的能力,在推动神经科学研究方面表现出了独特的优势[18]。在该技术中,特定的神经元通过基因工程的手段在细胞膜上表达出光敏分子通道(光敏蛋白),例如ChR2、ICC2、eNpHR3.0、eBR等[8]。这些光敏蛋白受到可见光波长的光源照射后能调控钠离子、钙离子、氢离子、氯离子等的跨膜流动,达到刺激(去极化)或者抑制(过极化)该神经元的效果[19-21],如

光遗传刺激采用400~700 nm波长范围内的可见光照射光敏蛋白以调控神经活动[19]。然而,由于大脑对可见光有很强的吸收和散射作用,可见光在脑组织中的穿透深度有限。为了实现更可靠地对深脑区和大脑皮层神经元的光遗传刺激,需要有特定的器件将光传导至目标刺激点位附近。可植入式光学神经探针(此处和下文中的“探针”均指传导光的工程器件,而非遗传编码荧光探针中“探针”所指的如GCaMPs指示剂等的蛋白表达工具)能够对此提供解决方案,其开发是近年来的研究重点[4]。
由于可植入式神经探针会不可避免地损伤生物组织,因此,开发基于长波长或红外波段的光遗传探针对制备侵入性更小的光遗传刺激系统也具有重要意义。这是因为长波长的光具有更弱的光毒性[25],在脑组织中的散射程度低、穿透性强[2]。最早实现光遗传刺激的光敏蛋白ChR2[19]的工作中心波长约为473 nm[26],Lin等[27]研制了最佳工作波长在590 ~ 630 nm的ReaChR光敏蛋白,采用放置于皮外的LED进行照射,实现了对小鼠触须运动的控制。Inagaki等[28]借助ReaChR通过LED的体外照射实现了对果蝇社会性行为的调控。此外,在激活神经元的研究中也可采用Chrimson和ChrimsonR蛋白(工作中心波长约为590 nm)[29],在抑制神经元的研究中则可采用Jaws蛋白(工作中心波长为632 nm)[30]。红外波段的光具有更优的穿透性能,有助于刺激深脑区,然而目前尚未发现相应工作波长的光敏蛋白[31]。因此,当前利用红外波段光实现光遗传刺激需要借助上转换纳米粒子,使进入脑组织的红外光经过目标神经元附近的上转换材料转换为可见光,进而激活光敏蛋白[32-33]。
目前,为实现更可靠的光遗传刺激,普遍开展的仍然是以可植入式探针为刺激工具的研究。在性能上要求植入后的探针出射光能准确照射目标神经区域,并且达到刺激阈值,实现对局部神经元的刺激。
总体上,光遗传探针除了用于刺激大脑皮层和深部脑区,还能用于刺激脊髓中的神经元[34]或者心脏和肌肉等其他器官[35-37],研究人员相应也开发了光纤神经袖带植入物[38]、硬膜外光纤植入物[39]、透皮照明[35-36]等诸多方案,这些方案进一步丰富了光遗传探针的应用场景。总体上,光遗传探针在过去十几年的发展中已经取得一些成果,对于脑科学而言,更常用的几种可植入式探针类型包括基于光学纤维结构的光纤型神经探针、集成微型发光二极管(μ-LED)型神经探针以及基于集成光学的神经探针等[40-45]。从临床应用的表现看,现已借助这些光遗传探针,缓解了患有帕金森症[23]、抑郁症[46]、癫痫[47]等疾病的小鼠的症状,克制了可卡因成瘾小鼠的毒品依赖性[48],重建了一种失明小鼠眼球对光的敏感度[49]。而对于非人灵长类动物恒河猴[49],现已经成功开展刺激其额叶皮层神经元并观察到神经响应信号的实验。对于人类[50],也已经成功开展了针对视网膜神经节细胞的刺激实验。
对于基于硬质材料制备的传统神经探针,探针的杨氏模量大约在10 GPa量级,远大于神经组织的杨氏模量(1~10 kPa)。在手术植入时,硬质神经探针的力学性能失配会直接对神经组织造成伤害[51]。对于长期植入而言,动物的呼吸和血管循环会使脑组织在颅腔内产生数个微米乃至数十微米的移动,力学性能失配的探针无法跟随神经组织移动因而对神经组织产生切割,而动物的行为运动和探针通过牵引结构传递的外部振动也会加剧这种切割的发生,最终导致生物组织产生严重的免疫反应[52]。免疫反应会促使星形胶质细胞逐渐在探针表面聚集,最终形成星形胶质瘢痕,以绝缘层的形式包裹探针,致使器件性能退化,记录的信噪比和刺激效率降低,探针的使用寿命缩短[53-56]。除了生物环境对探针性能的折损,刚性神经探针自身也可能产生裂纹或分层[57],并且与神经组织之间的相对移动会导致光遗传刺激点位发生偏移[58],刺激效果及准确度降低。因此,生物兼容的柔性神经探针的开发,对于减少手术伤害、炎症反应,以及实现高精度、长期稳定的神经调制有着重要意义。选择合适的柔性材料是实现柔性探针的基础[59-61]。由于器件的硬度除了探针材料之外还取决于探针的形状,宽而薄的柔性探针即便在弯曲时也可能在沿波导截面宽的方向产生更强的硬度,引起组织的反应[62]。因此,制作小尺寸、结构紧凑的探针同样为柔性探针的实现提供了思路,另外也有利于缩减探针植入手术的创口大小。目前基于柔性材料制备的光遗传探针仍主要面向动物行为开展生物实验,调控了动物运动(方向、速度、偏好与厌恶的位置)[22,51,63]与社交[64]的情况,以证明相应柔性探针方案的可行性,未来有望进一步系统性地研究将其用于神经疾病治疗的方案。
本综述提供了近期光学神经探针发展的概况。第一部分分析了精准神经调制对于脑科学等研究的意义,首先比较介绍了常用的神经调控手段,其中光遗传刺激技术在时空精度分辨上具有优越性,进而提出发展光刺激探针的原因。然后进一步在探针种类中强调发展柔性探针的必要性。第二部分讲述了光遗传探针中实现光刺激功能的三类探针:光纤型探针、μ-LED探针、波导集成型探针,在每一类探针中特别探究了柔性探针的研制情况。第三部分对光遗传探针的其他功能及其实现方式进行了分析,如原位电生理记录等,着重探讨了柔性探针在实现各种功能上的思路。第四部分总结了全文对于几种光遗传神经探针及其功能和性能的讨论,明确了理想神经探针应该具有的特点,并将其和最新的研究成果相对应进行展望,强调了光遗传探针在脑科学研究领域的发展方向和重要作用。
2 光遗传探针发展现状
光在脑组织中的有效传输并且完成光遗传学刺激是一项颇具挑战的任务。它要求光遗传学刺激探针的出射光功率充足,即便刺激对象位于深脑区,也能够有效地照射光敏蛋白,同时也要求刺激系统具备最小化的组织侵入性和热损伤,并与生物体的特性兼容。此外,还要求探针具备高空间、高时间分辨率的光遗传学刺激能力和神经电信号记录能力,以匹配单个神经元的尺寸与神经响应信号的时间[65]。为此,研究人员首先研制了光纤型探针,使植入生物体内的光纤能将光导引至需要照射的位点。为了试图解决光纤牵引与复杂庞大的外部光源的问题,他们进一步研制了能够实现无线工作模式的μ-LED型探针。随着微纳加工工艺的进步,近来基于集成光学的波导集成型探针也飞速发展,通过在微纳尺度进行元件设计,达到减小探针出射光斑尺寸,提升出光可控性的目的。在材料方面,这几类探针也分别引入了柔性材料进行探索,三种探针的生物兼容性得以提升。这些研究优化了光遗传探针的光刺激功能。
2.1 光纤型探针
光纤是进行体内光遗传研究的重要手段之一。首次成功的小鼠体内行为刺激实验便是用光纤实现的,由Deisseroth团队于2007年开展,如

为了刺激更多的脑部区域,各研究组分别开发了不同的方案。2008年Xu等[67]把光纤阵列和二维LED阵列耦合,将每一根光纤都与一个LED相连,通过控制LED的亮灭在出光口形成不同的照明图案。Royer等[68]于2010年设计了多柄探针,柄尖端集成了金属电极用于记录神经响应信号,每个柄上方分别承载了一根单模光纤。此外,在原有设计基础上,该团队尝试在各探针柄下方开槽内嵌光纤,以提升探针的出光位点数目,如

光纤型探针系统中,目标动物受限于光纤的牵引无法自由活动,这会为生物实验带来不便并且也可能影响实验结果的可靠程度。此外,动物在携带整个装置移动的过程容易导致光纤产生打结的问题,进而导致光纤断裂[72]。Klorig等[73]于2014年引入的光纤换向器能够解决这一问题。作为一个可旋转的关节它能保证小鼠的自由活动,同时也不会影响光传输的质量。虽然引入光纤换向器对光纤型探针系统有一定改善,但本质上这一类探针系统仍普遍存在一些问题:连接庞大复杂的外部光源结构让小鼠移动不便、通道数难以提升、出光光斑大难以实现单细胞刺激、出光光路不可控等。
并且,如上方案中均采用硬质光纤,其力学性能和脑组织并不匹配,平均杨氏模量(大约10 GPa)比脑组织(大约1 kPa)大6个数量级,植入时会对脑组织造成严重损伤[51]。而对于术后的长期阶段而言,因为大脑在颅腔内有一定的活动,探针和脑组织界面产生错切会导致炎症的发生。柔性探针有更接近脑组织的杨氏模量,在慢性植入阶段能够随着脑组织的微小移动而移动,相应制备柔性光纤探针的方案可以应对这一问题[63]。早期许多研究利用常用聚合物材料柔软的特点,开发了初步的柔性光纤型探针,但这些方案对柔性的提升并不显著,仍应采用其他材料来提升探针与脑组织的力学匹配程度[74-75]。2018年Wang等[76]以海藻酸钠-聚丙烯酰胺(PAAm)为主要原材料制作了水凝胶光纤探针,如

柔韧度高的材料植入后虽然对生物组织的损伤比硬质探针小,但其植入过程本身面临更多挑战。因为单独的柔性探针并不容易实现精准地植入,若辅以刚性的导引结构,从某种程度上讲反而会再度对脑组织造成损伤。2022年Zhou等[63]开发了以蚕丝为光纤材料的探针,如

总体上,当前实现柔性光纤型探针的关键点集中在材料的选取方面,包括本身具有更大柔性的材料,以及植入前后发生性变的材料。柔性探针通常面临植入困难的问题,除了会变性的蚕丝材料能够提供一种解决方案,寻找体内可降解的硬质材料辅助柔性探针的植入也是可行的,具体通过将柔性探针附着于特殊的硬质材料上,或者以硬质材料作为包层,保证探针植入时的刚度需求[78]。另外,大小和形状的设计常见于柔性电学器件,借鉴其中的思路也有助于提升光纤型探针的柔韧度[79]。即便柔性光纤型探针造成的脑组织损伤小,但依然面临除炎症反应之外硬质光纤型探针系统所造成的各种常见问题,仍需要开发其他类型的光遗传探针。
2.2 μ-LED型探针
第二类常见的光学神经探针使用μ-LED作为光源。这类神经探针通过植入μ-LED至特定神经区域附近并提供特定波长的光信号,实现光遗传刺激或者其他神经调制效果[44]。此类方案用低功耗的μ-LED光源取代了复杂庞大的外部光源,配合无线工作模式能够避免光纤对生物实验对象的牵引,这是因为电子驱动的LED适合与无线通信设备集成,使探针系统成为独立的组件,完全或部分植入生物体后保证了动物的自由活动[80]。
2010年,Grossman等[81]首次用自行制作的高功率μ-LED阵列进行了体外实验,实现了高密度高空间分辨率的神经调制。但这种相对简单的设计无法刺激深部脑区,单纯的平板面阵结构也不方便集成如原位电生理记录电极等其他结构。并且这种设计LED密度大,可能诱发产热问题。为了能够刺激大脑深部,μ-LED型探针有两种普遍形式[80]。其一是在μ-LED前端连接一小段光纤(波导)[82]或者光电极[83-84],如

柔性μ-LED型探针在设计思路上与光纤型探针不同,在不引入小段波导结构时,它引入的柔性材料主要作为承载μ-LED光源的基底,柔性材料本身不传输光,因此无须关注柔性材料的光学传输能力。目前常见的柔性μ-LED型探针结构以生物兼容的材料制备柔性衬底,在其上方集成μ-LED和其他光学与电学的结构[45]。常用的柔性衬底制备材料包括PDMS、SU-8、聚酰亚胺(PI)、Parylene C、环氧树脂等[87-88]。Kim等[22]开发了基于聚酯纤维的多功能柔性神经探针,如

特殊结构设计的引入,也为克服μ-LED的产热效应,提升出光效率提供了解决方案。Xie等[93]在μ-LED探针上引入了和柔性衬底集成兼容的超薄钻石薄膜结构,利用了钻石薄膜高达700~2200 W/(m·K)的导热系数,增强了μ-LED型探针的散热性能。Klein等[94]从减小μ-LED出射光发散的角度提高探针工作效率。他们采用先筑模后浇注再转移的方法,在聚合物衬底的μ-LED神经探针上增加了PDMS微透镜阵列,使神经探针在空气、水中的整体光采集效率分别提升了115%和83%,出射光峰值强度分别提升了145%和95%。Liu等[95]研发了一种生物兼容的光学滤波薄膜,它在420~550 nm区间内的反射效率约为99%,可以通过转印的方式与μ-LED(470 nm波长)背面结合,用于增强μ-LED的出光强度。这些设计均益于提升μ-LED型神经探针的出光效率,降低驱动μ-LED所需的电流强度,缓解μ-LED的产热问题。
柔性μ-LED型神经探针的发展非常迅速,已被广泛应用于各类光遗传神经调制实验中,总体的进展包括:开发了一体化的加工技术方案;提出了产热问题的多种解决策略;增加了探针的刺激通道数目;开发了双波长出射的功能等。但是对于μ-LED型神经探针而言,未来的发展依旧存在一定的挑战。由于μ-LED结构出光的发散性,限制了其进行高精度神经调制的能力,虽然为增大出光精度μ-LED的尺寸能够缩至10~20 μm,但尺寸减小的同时会损失出光效率,并且会引起严重的产热问题[96-97]。因此,开发长期安全稳定的、有着高精度刺激能力的柔性、小尺寸μ-LED型神经探针有望实现高时空分辨的光遗传刺激。
2.3 波导集成型探针
波导集成型探针的飞速发展得益于微纳加工工艺的不断进步,相比前两种类型的探针,它依赖微纳光学元件,对光的调控灵活性高。通过设计出光结构,探针出射的小尺寸光斑易于匹配单细胞的尺寸,达到单细胞精度的刺激。探针通过引入如Y型分束器、定向耦合器、多模干涉耦合器(MMI)等元件实现光的分束、合束,仅依赖单个光源便可实现多个出光位点的多通道刺激。在此基础上安装光开关器件,每路出射光会具有独立的可控性。探针引入响应随波长变化的光学相控阵(OPA)型器件,能够使出射光发生偏转,扩大了探针的有效刺激范围。另外,波导集成型探针的光从位于探针后端的光源耦合进入每根波导,这些光源保持在组织之外,避免了对组织或者照明过程产生影响。
2010年,Cho等[98]通过采用SU-8波导,首次在装有16个铱电极的探针柄上实现了单通道的波导集成型探针。但该探针未集成波导之外的微纳光学元件,硅基探针柄上仅集成了一根光刺激波导。为了增强出光可控性,波导集成型探针需要引入一系列微纳光学的单元器件。Son等[99]引入Y型分束器设计了基于SU-8波导的4通道硅基光遗传探针,如

除了在同一波长下实现多路分光,借助元件和波长之间固有的响应,可建立光波长和出光通道之间的对应关系,实现多通道的神经探针。Lanzio等[105]引入了无源微环谐振器结构,如
常规光栅耦合器具有特定的工作波长,出射光束方向恒定,为了从光束转向的角度提升探针出光的可控性,需要引入OPA辅助出光。OPA通过调制入射波长,进而调制OPA不同通道间出射光的相位差,表现为在某一个维度上远场出射光束角度的变化。2019年,Sacher等[106]报道了第一个利用氮化硅OPA作为微尺度光发射器的可植入式光束转向神经探针,如

上述探针中以SU-8为波导材料的几种设计,虽然加工便捷,但由于材料折射率不高,所制备的器件尺寸较大,不利于提升集成度。相比较而言,氮化硅在可见光波段透明,传输损耗低,并且可以使用CMOS兼容的标准沉积和蚀刻工艺来制造[104],作为波导集成型探针的材料更具优越性。
尽管上述设计在提升通道数、减小探针尺寸、减小出射光斑大小、增强光束可控性等方面有许多进展,但如上探针均采用硬质硅基衬底,在植入时虽然方便精准定位,却会对生物体自身造成伤害。类似于光纤型与μ-LED型探针,需要开发相应的柔性波导集成型探针来进一步拓宽应用前景。虽然相关研究起步较晚,但柔性波导集成型探针仍可借鉴其他两类光遗传探针的设计思路,甚至电学柔性神经探针的选材和结构。
Nizamoglu等[111]研制的基于左旋聚乳酸生物高聚物的梳状平板波导可以用于光化学组织黏合,成功在深层组织引起了光化学反应。虽然并不用于光遗传刺激,但这种柔性波导也为光遗传探针提供了参考。微纳尺度的光波导结构和光纤的制作不同,依托于硬质的衬底形成图案,柔性波导的图案同理。对此,制作波导集成型探针的主要思路是在硬质衬底上增加一层释放层(或称牺牲层),柔性波导附着在这一层结构上,再通过相应手段去除释放层,将柔性波导结构分离出来。2020年,Reddy等[112]以氧化硅为释放层制备了一种基于生物相容材料Parylene C(1.5~4 GPa)和PDMS(1.32~2.97 MPa)的柔性波导集成型探针,如
柔性波导集成型探针为了满足植入时的刚度需求,既可采用植入套管[116]或者利用包覆生物可吸收的加强剂[117],也可在植入前将波导前端刻蚀成尖锐状[118],或者采用另外的辅助支架等。
目前关于柔性波导集成型光遗传探针的研究较少,存在探针光刺激功能性不强、探针尺寸偏大的问题。并且当前研究多停留在技术验证阶段,基本未开展相应的生物实验。柔性探针中也未囊括硬质探针中功能多样的单元器件,导致探针缺乏出光光束可控性。因此,柔性波导集成型探针的研究亟需新的设计、加工和应用方案。
3 多功能的光遗传探针
先进的光学神经探针除了实现基本的光刺激,通常集成了用于表征和测量神经活动、药物递送、无线供能和数据传输等结构,这些设计丰富了探针的功能,使生物实验更加便捷。本章节将从这一方面出发,介绍包含多个功能单元的光学神经探针,其中重点介绍柔性的多功能神经探针的应用和研究进展。
3.1 电生理记录功能
集成有电生理记录功能的神经探针适于探测神经元被激活的情况。在神经系统中,神经回路通过神经元之间的生物电信号来传递、处理生物信息[119],这些生物电信号通过神经元上跨膜蛋白通道中的离子交换来实现[120]。光遗传刺激可以诱发神经元发生这种离子交换也即产生生物电信号,记录这些生物电信号是记录神经活动的重要方法之一,它也是光遗传刺激实验是否成功的证明之一[121]。单独引入的电极阵列记录探针虽然可以观测这些电信号,但是,此类方案通常伴随更大的植入创口。并且电记录探针的植入位置是否准确也值得考虑,尤其对于在光遗传探针后二次手术植入的电记录探针而言,只有将它植入在目标细胞旁才有可能探测到信号。直接在光学神经探针上集成电生理记录电极避免了这些问题。对于硬质探针而言,这种集成方式多在硬质的探针柄上排布电极,如

柔性探针的记录电极相应以柔性的衬底做支撑,例如上文中基于蚕丝材料的柔性光纤型探针,是基于前期Shi等[63,124]对柔性共形电子器件的探索。其记录电极以柔性PI为衬底,附着在蚕丝光纤上,如
事实上,光刺激有可能会对电记录造成一定干扰,在这种记录过程中若光照射到电极,则会诱发如光电效应等各种电伪影[125]。在波导集成性探针中引入光栅或微反射镜结构有利于规避这一问题,使出光方向垂直于探针柄。上文中Reddy等[112]设计的PDMS/Parylene C柔性探针便采用了这种方案提高电生理记录的准确程度,如
另外,发展更成熟的柔性神经电极阵列也为柔性光遗传探针集成电生理记录功能提供了重要参考。Xie等[127]开发的网格状大孔结构的三维神经电极阵列具有极强的生物兼容性,弯曲刚度比硅基电极阵列低4~7个量级[128]。在长期植入的过程中神经元会与电极阵列融为一体,形成稳定的接触界面。Luan等[129-130]研制了电极沿探针纵向分布的超柔性纳米电子线神经探针,其薄而微小,不易触发人体的免疫反应,并且纳米电子线也具有机械顺应性,易于实现与神经组织的无缝隙集成。Guan等[131]发明了一种自组装式的柔性神经流苏电极,先将流苏浸入聚乙二醇形成复合细丝,保护植入过程中脑组织不受电极的损伤,在植入后,聚乙二醇会在脑内降解。该方案相比前两者更便于提升通道数,并且该探针在长期实验中并未引起明显的神经元密度损失。这些方案主要依托于器件薄的结构[132]而非柔软的材料来实现柔性的神经电极阵列,和柔性光遗传探针的结合亟待进一步探索。其中,Guan等[131]已经开展了将流苏电极直接黏附在硬质光纤型光遗传探针表面做原位电生理记录的实验。
3.2 生物或化学递送功能
生物或化学递送功能有助于开展生物实验,通常采用基于MEMS的微流控通道和探针结构集成。动物活体光遗传刺激实验的手术通常分两次开展,第一次需要将病毒载体注射到特定的脑区,使特定的神经元表达光敏蛋白,第二次将光遗传探针植入动物脑内,使探针刺激选定的神经元。由于手术的分批次开展,一方面会造成二次创伤,另一方面必然会存在两次手术靶向偏差的可能,也即光遗传探针未能成功照射表达了光敏蛋白的神经元。药物递送的主要递送对象是辅助实验的药剂,也会面临同样的问题。因此,需要将实现这类功能的通道集成至光遗传探针上。
对于光纤型探针而言,多用热拉法将微流控通道集成入光纤端面的多芯结构中。Canales等[133]制作的环烯烃共聚物-聚碳酸酯柔性多功能光纤型探针中,保留出的中空通道即为微流控通道,如

除了微流控通道,具有生物兼容性和水溶性的丝纤维蛋白包覆载体病毒后再附着到植入探针的前端,也可在植入手术后将病毒于预定位置释放出来[136]。Zou等[137]研究了类似的具有生物兼容性和生物可降解性的熔融聚乙二醇。先将探针蘸入混有病毒载体的熔融聚乙二醇再拉出到空气中,带有病毒的聚乙二醇被探针中的柔性微电极细丝捕获并迅速固化,植入后再于生物体内降解释放病毒。这两种方案在植入前后的硬度以及形态变化有利于辅助柔性光遗传探针的植入。
3.3 无线供能和数据传输
无线工作模式的探针增添了器件整体的便携性,避免了牵引结构对装置、生物体以及实验结果造成的影响,通常采用感应的电学结构驱动探针上包括光源在内的有源器件。但无线操控目前主要针对μ-LED型神经探针,通过在器件上集成小型无线电频率共振模块、控制芯片和蓝牙模块等,即可实现整体器件的无线供能、控制和数据传输等功能[138-140]。

电生理记录电极的集成思路对集成探测其他物理量的传感器有借鉴意义。如Kim等[22]开发的多层结构的柔性探针,除了用于电生理记录的一层结构,还包含微型无机光电探测器,可用在脑组织深部实时监测μ-LED的出光情况,以及温度传感器,以接近1 mK的精度确定局部加热的程度,预防探针可能造成的组织热损伤。
总体上,除了追求更高性能的光刺激,针对光遗传探针附加功能的诸多研究丰富了光遗传探针的整个技术体系,为将来光遗传探针走出实验室实现产业化奠定了一定基础。
4 结束语
本文从光遗传学的背景切入,对光遗传探针的发展现状进行了介绍,首先对光纤型、μ-LED型与波导集成型三大探针类别下的多种设计进行了讨论,具体明确了:光纤型结构简单,但实验生物体与光源或其他外部复杂庞大的结构间存在牵引问题;μ-LED型采用无线模式后可以避免光纤牵引,但此类探针存在工作效率低、产热与加工工艺复杂等问题;波导集成型探针相比其他两者在光路可控性方面与实现单细胞刺激层面有显著的优势,也依然具有更多的发展空间。为减少生物组织损伤,针对这三类探针开发柔性探针具有必要性。对于柔性光纤型探针而言,通过热拉法集成多功能通道非常便捷,但却并不利于通道数的提升。并且为了减小光在脑组织中的损耗,柔性光纤型探针对材料折射率的要求更严格。对于柔性μ-LED型探针而言,层式结构非常普遍,它不必考虑材料导光的性能,但也面临柔性材料导热性差加剧LED的产热问题。对于柔性波导集成型探针而言,尚未有成熟的研究将硬质衬底上用于分光合光等的微纳光学元件引入柔性的光路中,相关研究亟待开展。本文还讨论了光遗传探针除光刺激功能之外的其他功能的集成方案。在多功能柔性探针的实现上,不仅介绍了特殊的柔性结构设计,也凸显了三类探针与柔性电子、生物兼容的性变材料,以及天线、控制芯片、蓝牙模块等更多其他领域技术的紧密联系。
对于目前的发展情况而言,三类探针各有利弊,我们设想未来会有一种侵害性更小的光遗传探针的设计,能实现多光谱、多通道、多目标、多功能的光学神经接口,实现无线、实时、双向和同时问询活体动物神经信号的工作模式,并且探针具有理想的系统稳定性和生物相容性[64-65,142]。现有的许多技术进展可以支持这一愿景,下面将对此进行简要讨论:
1)多光谱与双向。Li等[143]开发的三色堆叠式μ-LED在现有的双色探针基础上提供了更多波长的出光,为实现多光谱探针奠定了基础,该光源不仅能做到双向刺激,还为选用多种不同响应波长的光敏通道蛋白创造了可能。
2)多通道与无线。波导集成型探针非常利于实现多通道的光遗传学刺激。现有的芯片级集成光子回路的研究有利于为器件的设计思路提供参考[144-146]。若在波导集成型探针中不采用光纤耦合而将其光源更改为LED,则有望实现无线的工作模式,但此方案中光源和波导耦合效率低的问题仍待解决[72],具体的无线供能与数传模块也尚需进一步探索。
3)多目标。控制光束转向从而变换刺激目标的功能已经在波导集成型探针上得到了初步验证,针对光学相控阵的研究有利于实现探针发射光束的转向。其中,我们应着力解决旁瓣抑制问题,以提高刺激的空间分辨率[147]。此外,在变换刺激目标或者变换导通光路的过程中仍需要避免引入外部的机械结构,微纳尺度的有源光开关器件更适用于片上的一体化集成。
4)多功能与实时问询。多功能的实现是一项交融各个学科与领域的任务,现有的各种形式的探针在集成微电极与微流控通道方面,以及μ-LED型探针在集成无线通信模块方面已经取得初步进展。如何保证电记录器件在探针上尤其在柔性衬底上的性能,同时不影响光传输的效果[124],如何保证数据的实时传输、通信模块的准确收发以及系统的反馈刺激等需要开展更多专项的研究。此外,柔性电子器件中实现电刺激的相关设计也具有借鉴意义[148]。
5)系统稳定与生物相容。探针的材料选择是研究要点之一,也即考察材料是否具有柔性、生物兼容性、生物可降解性等特质。其中,借助植入前后有硬度变化的性变材料,实现从植入到刺激再到自然降解只需一次手术的侵害性相对较小,为后续探针的开发提供了重要参考[63,111]。对于光传输而言,材料对光场的限制能力也需要进行分析,为了实现脑深部刺激,要求光在探针中传输时损耗小,出射光的光功率密度达到开启光敏蛋白的阈值。因此,传导光的光纤和波导需要选用高折射率对比度的芯层和包层,通常无机材料波导相较于聚合物折射率对比度更大,有助于实现尺寸更小、结构更紧凑的光遗传探针[149-151]。近年来以氧化钛为材料制备了柔性热光开关[152-153],采用GeO2为牺牲层(用水来腐蚀去除)制备了高质量的柔性氮化硅薄膜[154],这些研究为开发基于无机材料的柔性波导集成型探针奠定了基础。另外,近年来的新型传感材料MXene因其具有出色的光学、电学、热学、力学性能并且与生物体兼容,也有望用于柔性光遗传探针的制备[155]。
6)小侵害性。若选用近红外波段的光搭配相应的光敏蛋白则有望消除上述讨论中的植入式探针对生物体的影响,因为近红外波段的光穿透深度更大,无需光学结构便能够传导至大脑深处。但目前尚未发现相应响应波长的光敏蛋白,若仍然选用可见光波段响应的光敏蛋白,则需要在目标刺激位点附近借助上转换纳米粒子将红外光转换为可见光,用可见光刺激神经元[156-157]。
如上讨论仅为实现一个功能和性能更加强大的光遗传探针而提出了初步的见解,许多工作和深入性的研究亟待完成。总而言之,光遗传技术为实现细胞的特异性刺激提供了一个可行的解决方案,但这离不开光遗传探针在多个方向的技术发展和进步,其中,研究和开发柔性光遗传探针是当前面临的重要挑战之一,此类探针的成功研制有助于实验以和生物体更兼容的方式开展。这对神经科学家探索理解神经编码、治疗神经疾病等诸多方面意义深远。
