不同光动力剂量对C6胶质瘤细胞胞内游离钙的影响

摘要
目的研究不同剂量光动力作用后C6胶质瘤细胞内游离钙 [Ca2+ ]i的变化,探讨钙超载在光动力诱导C6胶质瘤细胞坏死和凋亡过程中的作用. 方法常规培养 C6胶质瘤细胞,以不同光动力剂量处理对数生长期的实验组细胞后,应用Fluo- 3/AM为细胞内钙离子荧光指示剂,激光共聚焦显微镜测定各组 C6胶质瘤细胞内游离钙的变化;MTT法检测各组细胞的死亡率. 结果光动力作用后 C6胶质瘤细胞内游离钙明显升高,平均胞内钙离子浓度和细胞死亡率随着光动力剂量增加而增大.结论光动力作用后 C6细胞钙超载在光动力诱导肿瘤细胞坏死和凋亡过程中起重要作用.
